Defining the HIV Capsid Binding Site of Nucleoporin 153

Summary: This study comprehensively investigated the effect of mutating the amino acids of NUP153 on HIV capsid interaction using a multidisciplinary approach. Computational and experimental techniques were combined to reveal that only six amino acids of NUP153 specifically interacted with the HIV capsid. The simplicity of this interaction motif suggested the possibility of other FG-containing motifs interacting with the HIV-1 capsid. Furthermore, it was predicted that naturally occurring polymorphisms in human and nonhuman primates could disrupt NUP153 interaction with capsid, potentially providing protection against HIV-1 infection.
