Biological, toxicological and molecular docking evaluations of isoxazoline-thiazolidine-2,4-dione analogues as new class of anti-hyperglycemic agents

Summary: In this study, three new isoxazoline-thiazolidine-2,4-dione derivatives were synthesized and assessed for their inhibitory activities against alpha-amylase and alpha-glucosidase. The compounds exhibited potent inhibitory effects on both enzymes and showed no cytotoxicity in in vitro and in vivo evaluations. Computational analysis confirmed the molecular binding properties of the compounds with the active sites of the enzymes. These findings highlight the potential of the synthesized derivatives in controlling hyperglycemia and type 2 diabetes mellitus without causing hepatic toxicity.

Antiproliferative activity and molecular docking studies of new 4-oxothiazolidin-5-ylidene acetate derivatives containing guanylhydrazone moiety

Summary: A series of new compounds containing guanylhydrazone moiety were designed and synthesized. The synthesized compounds exhibited good antiproliferative activity, especially against human breast adenocarcinoma cells. Some of these compounds showed selectivity against healthy cells. Molecular docking studies revealed the interaction between the most active compound and cancer-related proteins.

Inhibitory Effect of Polyphenols from the Whole Green Jackfruit Flour against α-Glucosidase, α-Amylase, Aldose Reductase and Glycation at Multiple Stages and Their Interaction: Inhibition Kinetics and Molecular Simulations

Summary: This study investigated the antidiabetic potential of unripe jackfruit and identified two polyphenols with strong antihyperglycemic activity. The polyphenols were found to inhibit key enzymes involved in glucose metabolism and protein glycation through competitive and non-competitive mechanisms. In vitro experiments also confirmed the inhibitory effects on protein glycation. These findings suggest that the phenolic compounds in unripe jackfruit play a role in its antidiabetic properties.

Benzotriazole-oxadiazole hybrid Compounds: Synthesis, anticancer Activity, molecular docking and ADME profiling studies

The ultrasound method significantly increases reaction yields by reducing the reaction time. The synthesized compounds exhibit promising anticancer activity against PANC-1 cell line. Compound 4d shows the highest binding score for the active site of both proteins in molecular docking studies.

Determination of biological studies and molecular docking calculations of isatin-thiosemicarbazone hybrid compounds

Summary: This study reports the design, synthesis, structural elucidation, and investigation of cytotoxic and antimicrobial activity, as well as the inhibition effects on butyrylcholinesterase (BChE) and acetylcholinesterase (AChE) enzymes of isatinthiosemicarbazone hybrid compounds. The most potent inhibitor of both AChE and BChE was compound 13, and compound 8 exhibited the strongest cytotoxic activity on breast cancer cell lines. The hybridization of isatin and thiosemicarbazone skeleton played an essential role in the inhibition of enzymes and cytotoxic activity.
