Mechanisms of stearoyl CoA desaturase inhibitor sensitivity and acquired resistance in cancer

Nicole Oatman et al.

Summary: In glioblastoma, some patients have low levels of SCD expression, leading to the evolution of cell lines that can survive independent of SCD, despite unknown mechanisms. Cell lines that escape genetic and epigenetic alterations become highly dependent on SCD for survival. Resistance to SCD inhibition in SCD-dependent lines is mediated through a previously unknown FOSB mechanism, which can be blunted to extend survival in tumor-bearing mice.

Lipotoxic Impairment of Mitochondrial Function in β-Cells: A Review

Axel Roemer et al.

Summary: Lipotoxicity is a major contributor to the development of type 2 diabetes by promoting mitochondrial dysfunction; it can lead to ATP deficiency, decreased beta-cell viability, but physiologically fatty acids are necessary for beta-cell function under proper conditions; cellular senescence is associated with lipotoxicity, and plant-based nutrients such as polyphenols may have protective effects against lipotoxicity and cellular senescence.

Molecular Mechanisms of Apoptosis Induction and Its Regulation by Fatty Acids in Pancreatic β-Cells

Jan Sramek et al.

Summary: Chronic exposure to high concentrations of fatty acids can lead to dysfunction and death of pancreatic beta-cells. Saturated fatty acids promote apoptosis, while unsaturated fatty acids have a protective effect. The involvement of ER stress, the mitochondrial pathway, and multiple signaling pathways in this process is suggested.

The pancreatic beta cell: an intricate relation between anatomical structure, the signalling mechanism of glucose-induced insulin secretion, the low antioxidative defence, the high vulnerability and sensitivity to diabetic stress

Sigurd Lenzen

Summary: Insulin biosynthesis occurs in beta cells of the pancreas, which are vulnerable to oxidative stress due to low expression of H2O2-inactivating enzymes. This vulnerability threatens the functionality of beta cells and increases the risk of developing a diabetic metabolic state.
