The hypotaurine-taurine pathway as an antioxidative mechanism in patients with acute liver failure

Takamasa Mizota et al.

Summary: This study found that in acute liver failure, there is a decrease in metabolites related to glycolysis and pentose phosphate pathways, as well as high-energy phosphornucleotides, while methionine and its metabolites are increased. Furthermore, the increase in hypotaurine and taurine coincides with a decrease in total glutathione, suggesting a potential role of the hypotaurine-taurine pathway in oxidative stress protection.

The management of transitional care of patients affected by phenylketonuria in Italy: Review and expert opinion

Biasucci Giacomo et al.

Summary: This study examines the management of the transition period for patients with phenylketonuria (PKU) and highlights the lack of sufficient healthcare assistance for adult patients. The authors propose a structured pathway for an efficient transition, including planning, team collaboration, and the appointment of a transition coordinator.

Analysis of S-Adenosylmethionine and S-Adenosylhomocysteine: Method Optimisation and Profiling in Healthy Adults upon Short-Term Dietary Intervention

Aida Corrillero Bravo et al.

Summary: S-adenosylmethionine (SAM) is essential for methyl transfer reactions. SAM is produced de novo via the methionine cycle. The demethylation of SAM produces S-adenosylhomocysteine (SAH), an inhibitor of methyltransferases. Measurement of SAM and SAH in plasma is valuable for diagnosing inborn errors of metabolism and assessing methyl group homeostasis.

Characterization of Choline Nutriture among Adults and Children with Phenylketonuria

Meriah S. Schoen et al.

Summary: This study compared choline consumption among individuals with phenylketonuria (PKU) to a reference sample and identified factors that may affect choline needs. The findings suggest that only a small percentage of PKU patients achieve adequate choline intake, and treatment methods can impact choline levels. Without fortified medical foods, choline and other related nutrient intake may be reduced. Regular monitoring of choline intake and increased fortification of medical foods could benefit this population.

Oxidative stress in phenylketonuria-evidence from human studies and animal models, and possible implications for redox signaling

Vanessa Trindade Bortoluzzi et al.

Summary: Phenylketonuria (PKU), a common inborn error of amino acid metabolism, can lead to severe neurological and psychiatric symptoms if not managed properly with early diagnosis and diet therapy. Oxidative stress is implicated in the pathogenesis of PKU, affecting metabolic pathways and potentially disturbing redox signaling, although further research is needed to fully understand the underlying neuropathophysiology.

Differences of Phenylalanine Concentrations in Dried Blood Spots and in Plasma: Erythrocytes as a Neglected Component for This Observation

Dorothea Haas et al.

Summary: Monitoring phenylalanine concentrations is crucial for managing phenylketonuria. Variations in Phe concentrations between different blood sample types have been observed, with significantly lower Phe levels in erythrocytes compared to plasma. Reliable conversion of dried blood spots into plasma concentrations is essential for therapy recommendations.

Inhibition of metabotropic glutamate receptor III facilitates sensitization to alkylating chemotherapeutics in glioblastoma

Julian P. Maier et al.

Summary: Glioblastoma demonstrates dynamic responses to its microenvironment and resistance to treatment. Glutamate signaling, particularly through the metabotropic receptor 3 (GRM3), plays a crucial role in modulating chemoresistance and transcriptional profile of GBM cells. Inhibition of GRM3 receptor enhances sensitivity of GBM to chemotherapy, providing potential therapeutic strategies for GBM treatment.

A single-cell type transcriptomics map of human tissues

Max Karlsson et al.

Summary: Advances in molecular profiling have enabled the creation of high-resolution single-cell type maps of human tissues, combining single-cell transcriptomics analysis with spatial antibody-based protein profiling. This technology allows researchers to explore the expression of human protein-coding genes and determine the number of elevated genes in different cell types, facilitating comparisons with bulk transcriptomics data. The analysis reveals distinct expression clusters corresponding to cell types with similar functions within and between organs.

Serine catabolism generates liver NADPH and supports hepatic lipogenesis

Zhaoyue Zhang et al.

Summary: The liver relies on a unique cytosolic serine catabolic pathway for NADPH generation, while adipose tissue uses the pentose phosphate pathway. Acetyl-CoA for lipogenesis in liver is derived from acetate and lactate, whereas in adipose tissue glucose is the main substrate. Despite using the same enzymes, different substrates are utilized, highlighting the potential for tissue-specific pharmacological interventions.
