ZEB1 induces ROS generation through directly promoting MCT4 transcription to facilitate breast cancer

Xiao Han et al.

Summary: This study investigated the correlation between the transcription factor ZEB1 and the metabolism-related gene MCT4 in breast cancer. It was found that ZEB1 directly binds to the promoter region of MCT4 and enhances its expression. Furthermore, the synergy between ZEB1 and MCT4 promotes breast cancer growth through the generation of reactive oxygen species (ROS) and autophagy. The findings of this study provide a theoretical basis for targeting metabolism in breast cancer therapeutics.

Single-cell resolved imaging reveals intra-tumor heterogeneity in glycolysis, transitions between metabolic states, and their regulatory mechanisms

Hiroshi Kondo et al.

Summary: The study tracks the transition and heritability of metabolic states in single PIK3CA mutant breast cancer cells, identifying non-genetic glycolytic heterogeneity. It reveals distinct subpopulations of cells with regulated glycolytic and mitochondrial metabolism by PI3K signaling, bromodomain activity, and cell crowding effects, showing how targeting these pathways affects specific subpopulations of cancer cells.

Fatty acid oxidation and autophagy promote endoxifen resistance and counter the effect of AKT inhibition in ER-positive breast cancer cells

Lei Duan et al.

Summary: The study revealed increased activation of AKT and AMPK in TAM and endoxifen-resistant cells, accompanied by metabolic reprogramming and increased autophagy. Concurrent inhibition of AKT and FAO/autophagy is crucial for sensitizing resistant cells to endoxifen.
