Binding Interactions between Receptor-Binding Domain of Spike Protein and Human Angiotensin Converting Enzyme-2 in Omicron Variant

Bahaa Jawad et al.

Summary: The emergence of the new SARS-CoV-2 Omicron variant has worsened the COVID-19 pandemic due to its large number of mutations, particularly in the receptor-binding domain (RBD), making it highly contagious and/or resistant to vaccines. Through molecular dynamics simulations, it has been found that the Omicron variant's RBD binds to ACE2 more efficiently and tightly, resulting in increased infectivity and transmissibility compared to other strains. Some of the RBD mutations have been predicted to affect antibody neutralization either through conformational changes in the spike protein or by altering its surface charge distribution. Other mutations either decrease or increase ACE2 binding.

A Vaccine of SARS-CoV-2 S Protein RBD Induces Protective Immunity

Qiaoqiao Qu et al.

Summary: The fusion of RBD protein and 6HB has successfully produced a purified protein named RBD-6HB, which when combined with AL/CpG adjuvant can stimulate animals to produce high-level antibodies and establish an effective protective barrier against challenges. This highlights the importance of trimerized SARS-CoV-2 S protein RBD in designing vaccines and provides a rationale for developing a protective vaccine through the induction of antibodies against the RBD domain.

A Computational Approach to Evaluate the Combined Effect of SARS-CoV-2 RBD Mutations and ACE2 Receptor Genetic Variants on Infectivity: The COVID-19 Host-Pathogen Nexus

Dana Ashoor et al.

Summary: Understanding SARS-CoV-2 infectivity involves studying the interactions between the virus Spike protein and the ACE2 receptor, impacted by RBD mutations and ACE2 polymorphism. Analysis shows that the majority of RBD point mutations destabilize the virus-receptor complex and different ACE2 variants lead to varying molecular interaction patterns and complex stability. Considering both ACE2 variants and SARS-CoV-2 RBD mutations is crucial for evaluating the stability of the virus-receptor association and infectivity.
