Induced expression of miR-1250-5p exerts tumor suppressive role in triple-negative breast cancer cells

Summary: Triple-negative breast cancer (TNBC) is a highly aggressive subtype of breast cancer with relatively unknown pathogenesis. In this study, the researchers investigated the tumor-suppressive role of microRNA (miR)-1250-5p in TNBC cells through various experiments. They found that increased expression of miR-1250-5p in TNBC cells led to decreased cell viability, inhibited G1/S phase transition, induced apoptosis, and reduced epithelial-mesenchymal transition. The results suggest that miR-1250-5p could be a potential therapeutic target for TNBC treatment.

Natural Steroidal Lactone Induces G1/S Phase Cell Cycle Arrest and Intrinsic Apoptotic Pathway by Up-Regulating Tumor Suppressive miRNA in Triple-Negative Breast Cancer Cells

Summary: In this study, it was found that Withaferin A (WA) can exert an anticancer effect on triple-negative breast cancer cells by modulating the expression of miRNA. The results showed that WA up-regulated the expression of miR-181c-5p and down-regulated the expression of miR-1275, among other miRNAs. These miRNAs were found to affect the expression of cancer-associated genes and signaling pathways. Additionally, co-treatment of WA and miR-181c-5p mimic inhibited cell growth, induced apoptosis, and regulated the expression of genes involved in cell cycle and apoptosis.

The clinical relevance of these miRNAs was confirmed by survival analysis in breast cancer patients, showing poor overall survival and high expression levels in invasive breast cancer subgroups. Further validation in in vitro models demonstrated the downregulation of lead miRNA with phytochemical treatment.
