This paper discusses recent trends in developing Tacrine and Donepezil analogs and multifunctional hybrid ligands for treating AD.

Effect of metformin on intact mitochondria from liver and brain: Concept revisited

Summary: Metformin, an antihyperglycemic drug, has potential as a treatment for cardiovascular disease in individuals without diabetes. This study investigated the effect of metformin on intact mitochondria and found that it inhibits respiration and stimulates ROS release by complex I-based respiration. However, metformin had no effect on respiration rates and inhibited ROS release when mitochondria oxidize succinate or glycerol 3-phosphate. Additionally, metformin was found to weakly affect the active/deactive transition of mitochondrial complex I. These findings highlight the importance of metformin in regulating ROS production during the ischemia/reperfusion process.

Korean Red Pine (Pinus densiflora) Bark Extract Attenuates Aβ-Induced Cognitive Impairment by Regulating Cholinergic Dysfunction and Neuroinflammation

Summary: In this study, Korean red pine bark extract (KRPBE) demonstrated an anti-amnesic effect against amyloid beta1-42-induced neurotoxicity. KRPBE improved behavioral function, boosted the antioxidant and cholinergic system, ameliorated cerebral mitochondrial deficit, modulated neurotoxicity indicators and inflammatory cytokines, and regulated neuronal apoptosis-related markers. Therefore, KRPBE treatment has potential as a therapeutic approach for cognitive impairment associated with amyloid beta-induced neurotoxicity.

Metformin alleviates neurocognitive impairment in aging via activation of AMPK/BDNF/PI3K pathway

Summary: The therapeutic effects of metformin in d-galactose-induced aging were evaluated, and the potential molecular mechanisms responsible for its anti-aging effects were studied.
