Lipid-liquid crystals for 2 months controlled risperidone release: In-vitro evaluation and pharmacokinetics in rabbits

Summary: This study developed a drug delivery system based on lipid liquid crystal (LLC) for long-term delivery of risperidone to improve psychological treatment. The optimized LLC formulation achieved maximum release after 60 days using different ratios of phosphatidylcholine (PC) to sorbitol monooleate (PC: SMO), tween grade 80, and tocopherol acetate (TA). In vitro and ex vivo studies, pharmacokinetics, and histopathological examination in rabbits were conducted to compare the optimized LLC with Risperdal CONSTA (R). The results showed that the optimized LLC formulation had the highest drug release percentage (100%) and better pharmacokinetic potentials compared to Risperdal CONSTA (R).

The Impacts of PLGA/PEG Triblock Copolymers with Variable Molecu-lar Weights on the Sustained Release of Buprenorphine

Summary: This study focused on the preparation of a BP in-situ forming composite (ISFC) using different molecular weights of PLGA-PEG-PLGA triblock. The results demonstrated that ISFC 1500 exhibited better biocompatibility and suitable early initial burst reaction compared to ISFC 3000 and 4000, making it a potential candidate for the preparation of sustained-release BP formulations.

Optimization and in Vitro Evaluation of Injectable Sustained-Release of Levothyroxine Using PLGA-PEG-PLGA

Summary: In this study, a triblock polymer was synthesized using the melting method for controlled release of levothyroxine from in situ-forming gels. Increasing the concentration of triblock in the formulation significantly reduced the initial burst release of levothyroxine. The triblock showed thermosensitive properties and hydrogen bonding, leading to a smaller initial burst release compared to in situ forming implants.

Summary: This study demonstrates that the nanofibers coencapsulated with nTiO2 and MET@MSNs can prolong the in vitro expansion of human adipose-derived stem cells without inducing cellular senescence and aging. The platform enhances adhesion, metabolic activity, and proliferation rate of the cells, and leads to the upregulation of autophagy and stemness markers as well as the down-regulation of senescence marker. Moreover, it results in increased telomere length and telomerase activity in the cells cultured on the nanofibers compared to other types.
