Organic/Inorganic Self-Assembled Hybrid Nano-Architectures for Cancer Therapy Applications

Chengbin Yang et al.

Summary: The translation: The field of nanomedicine has advanced into clinical trials, but drug formulations for cancer treatment still face limitations due to biological barriers. To overcome these challenges, organic/inorganic hybrids have been developed to integrate the benefits of both types of nanomaterials through a self-assembly approach.

Succinimido-Ferrocidiphenol Complexed with Cyclodextrins Inhibits Glioblastoma Tumor Growth In Vitro and In Vivo without Noticeable Adverse Toxicity

Feten Najlaoui et al.

Summary: SuccFerr has remarkable antiproliferative effects and shows potential for clinical research. The water insolubility of SuccFerr is solved by complexation with RAMEssCDs, enabling in vivo experiments. The compound does not exhibit toxic effects on organs in vivo, and it can halt tumor progression in rats with xenografted tumors, possibly through an immunological defense response.

p722 ferrocifen loaded lipid nanocapsules improve survival of murine xenografted-melanoma via a potentiation of apoptosis and an activation of CD8+ T lymphocytes

Solene Topin-Ruiz et al.

Summary: The ferrocifen family represents a promising option for melanoma treatment, particularly when encapsulated in lipid nanocapsules with p722. This study showed that LNC-p722 significantly decreased melanoma cell viability and improved survival rates in mice models compared to anti-CTLA4 mAb therapy. Additionally, LNC-p722 was found to enhance the intrinsic apoptotic pathway and activate CD8(+) T lymphocytes, indicating a potential new therapeutic strategy for melanoma.

Multifaceted chemical behaviour of metallocene (M = Fe, Os) quinone methides. Their contribution to biology

Anne Vessieres et al.

Summary: Organometallic quinone methides (OM-QMs) exhibit unique chemical and biological properties when incorporated into specific substrates like ferrocifens, leading to disruption of cancer cell metabolism and cytotoxicity. Among these, ferrocifens show the highest toxicity within the iron, ruthenium, osmium triad, and the complexes can be utilized as imaging probes.

Gemcitabine lipid prodrug nanoparticles: Switching the lipid moiety and changing the fate in the bloodstream

Eleonore Coppens et al.

Summary: The study showed that inducing drug insertion into lipoproteins through chemical modification is a simple approach to achieve lipoprotein-mediated drug delivery. While the lipid moiety plays a key role in the interaction between the drug and lipoproteins, the overall physicochemical features of the nanoparticles must also be carefully considered.
