Ploidy Testing of Blastocoel Fluid for Screening May Be Technically Challenging and More Invasive Than That of Spent Cell Culture Media

Wenhao Shi et al.

Summary: This study demonstrates that spent cell culture media (SCM) has superior diagnostic performance compared to blastocoel fluid (BF) and is non-invasive for embryos.

The effects of the day of trophectoderm biopsy and blastocyst grade on the clinical and neonatal outcomes of preimplantation genetic testing-frozen embryo transfer cycles

Linjun Chen et al.

Summary: This study analyzed the impact of TE biopsy day and blastocyst grade on clinical and neonatal outcomes. The results showed that day 5 TE biopsy had higher rates of implantation and live births compared to day 6 TE biopsy. The miscarriage rate was lower for day 5 TE biopsy, but the difference was not statistically significant. Higher quality blastocysts achieved better implantation and live birth rates. Among good quality blastocysts, there was no significant difference between day 5 and day 6 TE biopsy rates. However, for fair and poor quality blastocysts, day 5 TE biopsy had significantly higher rates than day 6 TE biopsy. Neither blastocyst grade nor TE biopsy day significantly affected the miscarriage rate. Neonatal outcomes such as newborn sex, gestational age, preterm birth, birth weight, and low birth weight did not significantly differ between day 5 and day 6 TE biopsies. Both blastocyst grade and TE biopsy day should be considered together in preimplantation genetic testing-frozen embryo transfer.

Non-invasive preimplantation genetic testing for putative mosaic blastocysts: a pilot study

Xinyuan Li et al.

Summary: The study showed that using non-invasive preimplantation genetic testing (niPGT) on cell-free DNA in blastocyst culture medium can provide reliable diagnostic performance for putative mosaic blastocysts. This novel approach offers potential for reassessment of mosaic embryos and may be a minimally invasive option for maximizing the use of blastocysts. However, the study is limited by its small sample size, highlighting the need for further research to validate these findings.

ChromInst: A single cell sequencing technique to accomplish pre-implantation comprehensive chromosomal screening overnight

Fang-Fang Gao et al.

Summary: NGS has shown promising results in pre-implantation comprehensive chromosomal screening, improving the success rate of in vitro fertilization. The ChromInst protocol offers a concise and effective approach for single cell sequencing, allowing for timely support for fresh embryo implantation and accurate differentiation between normal and abnormal embryos.

Extended embryo culture is effective for patients of an advanced maternal age

R. Sainte-Rose et al.

Summary: The study found that blastocyst development rate is significantly reduced in advanced maternal age patients, with blastocysts showing growth retardation on day 5. While cell organization is unaffected by age, blastocysts with 'good' morphology in AMA patients have decreased implantation probability compared to 'average' morphology blastocysts in younger women. Although extended embryo culture is not harmful for AMA patients, it cannot compensate for the reduced implantation and live birth rates associated with maternal age.

Chromosome Screening of Human Preimplantation Embryos by Using Spent Culture Medium: Sample Collection and Chromosomal Ploidy Analysis

Jin Huang et al.

Summary: NICS technology may offer a solution to the problems in the prevailing PGT-A method, but its efficacy has been limited. A new study reports the full protocol of NICS, including culture medium sampling, whole genome amplification, and data analysis methods. Cryopreservation times in different embryo laboratories influence the choice of embryo culture medium collection methods.

A Non-invasive Chromosome Screening Strategy for Prioritizing in vitro Fertilization Embryos for Implantation

Li Chen et al.

Summary: The study validated the effectiveness of NICS testing in human embryos, which is comparable to PGT-TE and can be used to select embryos with normal ploidy for implantation. The scoring system provided by NICS allows for prioritization of embryos, and the addition of TE testing as a secondary validation significantly improves accuracy.
