Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Selective Targeting of Cancer Cells by Copper Ionophores: An Overview

Valentina Oliveri

Summary: Conventional cancer therapies face challenges due to off-target effects, leading to the need for new therapeutic agents with increased selectivity, reduced side effects, and the ability to overcome cancer cell resistance targeting cancer stem cells. Some copper ionophores show promise in inducing cuproptosis in cancer cells with greater selectivity.

LncRNA RP3-525N10.2-NFKB1-PROS1 triplet-mediated low PROS1 expression is an onco-immunological biomarker in low-grade gliomas: a pan-cancer analysis with experimental verification

Yujie Zhou et al.

Summary: This study demonstrates the significant role of PROS1 in low-grade gliomas, particularly its correlation with tumor immune infiltration. The long non-coding RNA RP3-525N10.2 may regulate the expression of PROS1, which has implications for the prognosis of low-grade gliomas.

Interactions between cancer cells and immune cells drive transitions to mesenchymal-like states in glioblastoma

Toshiro Hara et al.

Summary: The mesenchymal subtype of glioblastoma is believed to be influenced by both cancer cell-intrinsic alterations and external cellular interactions, with macrophages inducing a transition of glioblastoma cells into mesenchymal-like states. This transition is mediated by macrophage-derived oncostatin M (OSM) interacting with its receptors on glioblastoma cells, leading to the activation of STAT3. This study also reveals that mesenchymal-like glioblastoma states are associated with increased expression of a mesenchymal program in macrophages and enhanced cytotoxicity of T cells, suggesting potential therapeutic implications.

Inhibiting DNA-PK induces glioma stem cell differentiation and sensitizes glioblastoma to radiation in mice

Xiaoguang Fang et al.

Summary: This study demonstrates that DNA-PK regulates SOX2 stability through phosphorylation, contributing to the maintenance of Glioblastoma stem cells (GSCs). The findings suggest that DNA-PKcs-mediated phosphorylation of S251 is critical for SOX2 stabilization and GSC maintenance, highlighting the potential of DNA-PK as a therapeutic target in glioblastoma.
