TEMPO-Assisted Free-Radical-Initiated Peptide Sequencing Mass Spectrometry for Ubiquitin Ions: An Insight on the Gas-Phase Conformations

Summary: TEMPO-assisted FRIPS MS is used to analyze the gas-phase conformations of guanidinated ubiquitin. The study shows that thermal activation of UBT(Gu) ions produces radical protein backbone fragments, whereas CID results for UB(Gu) primarily show specific fragments. Radical transfer through space is observed in +5 and +6 UBT(Gu) ions. FRIPS MS analysis reveals that +5 charge state ubiquitin ions have a conformational resemblance to native ubiquitin in the central sequence region.

Summary: A newly designed TEMPO-FRIPS reagent was synthesized for single-step free radical-initiated peptide sequencing mass spectrometry (FRIPS MS), and it showed improved conjugation efficiency for target peptides compared to the previous version. The application of the new TEMPO reagent improved the duty cycle and has the potential to be used in proteomics studies, including phosphoproteomics.
