Discovery of a Cryptic Nitro Intermediate in the Biosynthesis of the 3-(trans-2′-Aminocyclopropyl)alanine Moiety of Belactosin A

Alicia Engelbrecht et al.

Summary: This study reveals the biosynthetic mechanism of Belactosin A by establishing a heterologous pathway expression and constructing gene deletion mutants. It is found that a cryptic nitrocyclopropylalanine intermediate is generated from L-lysine, and then converted to the corresponding amine through the reduction reaction mediated by the enzyme BelN.

Molecular Basis for Enzymatic Aziridine Formation via Sulfate Elimination

Sumire Kurosawa et al.

Summary: This study reports for the first time the molecular basis for aziridine ring formation. By investigating the biosynthesis of vazabitide A, which has a structure similar to azinomycin B, it was found that Vzb10/11 catalyzed the formation of the aziridine ring via sulfate elimination. Structural analysis revealed the working mechanism of these enzymes.

Stereodivergent Nitrocyclopropane Formation during Biosynthesis of Belactosins and Hormaomycins

Shotaro Shimo et al.

Summary: This study demonstrates that BelK and HrmI enzymes catalyze the N-oxygenation of L-lysine to generate 6-nitronorleucine, which is then cyclized by BelL and HrmJ enzymes to form the nitrocyclopropane ring with stereochemistry found in natural products. The study also reveals the inversion and retention of the configuration at C4 during the BelL and HrmJ reactions, providing insight into the unique strategy for stereocontrolled cyclopropane synthesis in nature.
