Versatile Types of Inorganic/Organic NIR-IIa/IIb Fluorophores: From Strategic Design toward Molecular Imaging and Theranostics

Summary: In vivo imaging in the second near-infrared window (NIR-II) has provided great opportunities for biomedical research and clinical applications. This review summarizes the design, properties, molecular imaging, and theranostics of inorganic/organic NIR-IIa/IIb fluorophores. The versatile applications and imaging modalities enabled by these advanced fluorescent properties are discussed, along with the challenges and perspectives for future clinical translation to boost the progress of NIR-IIa and NIR-IIb imaging technology.

Se-sensitized NIR hot band absorption photosensitizer for anti-Stokes excitation deep photodynamic therapy

Summary: This study presents a novel approach for constructing highly efficient phototherapeutics for low-power anti-Stokes light-triggered deep photodynamic therapy. By introducing selenium as a booster effector, in combination with hot band absorption luminophores, the photogenerated singlet oxygen capability was significantly enhanced at low power. PEGylation of the material further improved its tumor accumulation, leading to effective inhibition of deep-seated tumor progression.

Following clonal TP53 mutations, loss-of-heterozygosity events, and genome doubling, primary tumors undergo a period of transient genomic instability, with ongoing copy number evolution during expansion. Subcloned daughter cells demonstrate genomic rediversification, showing that triple-negative breast cancers continually evolve chromosome aberrations and maintain subclonal diversity during primary tumor growth.

Tailored Engineering of Novel Xanthonium Polymethine Dyes for Synergetic PDT and PTT Triggered by 1064 nm Laser toward Deep-Seated Tumors

Summary: In this study, five novel NIR-II dyes were successfully developed with intense absorption ranging from 890 to 1206 nm, strong singlet oxygen generation capability, and fluorescence imaging ability triggered by a single NIR-II light. Among them, BH 1024 showed the best performance in singlet oxygen generation, moderate photothermal heating, and considerable fluorescence under 1064 nm laser irradiation. Encapsulating BH 1024 into folate-functionalized polymer demonstrated a synergetic PDT/PTT effect in vitro and in vivo, eventually achieving solid tumors elimination under NIR-II fluorescence guide. This is the first attempt to explore and design small molecular dyes for NIR-II PDT or NIR-II PDT/PTT.
