Self-Assembled Metallacage with Second Near-Infrared Aggregation-Induced Emission for Enhanced Multimodal Theranostics

Summary: In this study, a prism-like metallacage CDTTP with efficient fluorescence emission in the second near-infrared (NIR-II) region was designed and constructed through the assembly of an aggregation-induced emission-active four-arm ligand with 90 Pt acceptors. C-DTTP exhibited the longest maximum emission wavelength (1005 nm) compared to previously reported SCCs and showed high photothermal conversion efficiency and superior reactive oxygen species generation behavior. In vitro and in vivo assessments demonstrated that the metallacage-loaded nanoparticles were capable of precise tumor diagnosis and complete tumor elimination through NIR-II fluorescence/photothermal dual imaging-guided photodynamic/photothermal synergistic therapy.

Molecular engineering of AIE luminogens for NIR-II/IIb bioimaging and surgical navigation of lymph nodes

Summary: This study conducted molecular engineering to design a near-infrared-II aggregation-induced emission luminogen with ultralong absorption wavelength for lymph node imaging and surgical navigation. Compared with the clinically used indocyanine green, the designed nanoparticles showed longer retention time, higher penetration depth, better signal-to-noise ratio, and greater photostability, enabling fast and accurate navigation of sentinel lymph nodes.

Summary: The study successfully constructed highly stable NIR-II AIEgen DPBTA-DPTQ nanoparticles using benzo[c]thiophene as an electron-rich and bulky donor/π-bridge, achieving acceptable fluorescence quantum yield and excellent photothermal conversion efficiency. DPBTA-DPTQ showed good biocompatibility and efficient photothermal therapeutic effects in in vitro tests, demonstrating potential for photothermal eradication of tumors in mouse models.
