However, the combination of directed evolution and computational design is bringing UPOs closer to practical applications.

Production of microhomogeneous glycopeptide by a mutated NGT according FuncLib with unique sugar as substrate

Summary: N-glycosylation is an important post-translational modification of proteins. A cytoplasmic soluble N-glycosyltransferase (NGT) in bacteria can transfer different sugars to proteins, leading to heterogeneity and complexity of glycopeptides. A mutant ApNGT-P1 with increased substrate specificity was obtained, which can be used for efficient production of glycosylated peptides or proteins with uniform glucose at glycosylation sites. This study also provides a feasible pathway for further research on improving donor substrate selectivity of NGTs.

Stabilization of the SARS-CoV-2 receptor binding domain by protein core redesign and deep mutational scanning

Summary: In this study, the researchers successfully stabilized the receptor binding domain of the SARS-CoV-2 Spike protein using a combination of deep mutational scanning and computational design. Their design maintained recognition by the receptor ACE2 and different monoclonal antibodies, and showed improved thermal stability and reduced proteolytic sensitivity compared to the original domain. This approach could be applied to stabilize a wide range of proteins without detailed knowledge of their active sites or binding epitopes.

Engineered Enzymes Enable Selective N-Alkylation of Pyrazoles With Simple Haloalkanes

Summary: Selective alkylation of pyrazoles using a catalyst-controlled enzymatic system allows for the efficient synthesis of important molecules with unprecedented regioselectivity. The use of a computational enzyme-library design tool enabled the conversion of a promiscuous enzyme into a small enzyme family of pyrazole-alkylating enzymes in one round of mutagenesis and screening. This enzymatic system demonstrates high selectivity and achieves different alkylations of pyrazoles with remarkable regiodivergence.
