Dual Nickel/Photoredox-Catalyzed Deaminative Cross-Coupling of Sterically Hindered Primary Amines

Julia R. Dorsheimer et al.

Summary: This study reports a new method to activate alpha-3 degrees amines for deaminative arylation by condensing with an electron-rich aldehyde and combining this reactivity with nickel metallaphotoredox to generate benzylic quaternary centers. The reaction is accelerated by added ammonium salts, which inhibit nickel black formation and accelerate the overall reaction rate. The study demonstrates a broad scope of coupling partners and shows an expedited synthesis of ALK2 inhibitors.

Mechanistic Insight into Metal Ion-Catalyzed Transamination

Robert J. Mayer et al.

Summary: Metal ions can catalyze transamination under biologically relevant conditions, with Cu2+, Ni2+, Co2+, and V5+ identified as the most active. These ions primarily accelerate the reaction by stabilizing key imine intermediates or increasing the acidity of the bound imine. Direct metal ion-catalyzed amino group transfer is highly favored even in the absence of cofactors or protein catalysts under biologically compatible reaction conditions.

Nickel-catalyzed deaminative Sonogashira coupling of alkylpyridinium salts enabled by NN2 pincer ligand

Xingjie Zhang et al.

Summary: This study presents a nickel-catalyzed deaminative Sonogashira coupling of alkylpyridinium salts to prepare alkyl-substituted alkynes efficiently. The development of a new easily accessible and bench-stable ligand enables alkyl amines to serve as alkylating agents in Sonogashira reactions with excellent yields.
