Simultaneous Visualization of Dual Intercellular Signal Transductions via SERS Imaging of Membrane Proteins Dimerization on Single Cells

Summary: A highly sensitive and specific surface-enhanced Raman scattering (SERS) strategy was proposed to simultaneously image dual homodimerizations of membrane proteins on single live cells. Networking of AuNPs-based dual-recognition probes (dual-RPs) and SERS tags via proximity ligation-assisted catalytic hairpin assembly (CHA) was used to enhance the SERS effect. This strategy provides a good method for high-sensitivity monitoring of membrane proteins dimerizations-based multiple intercellular signal transductions in a natural and complex cellular microenvironment.

Ligand-induced transmembrane conformational coupling in monomeric EGFR

Summary: In this study, the authors show that transmembrane conformational coupling is the first step in EGFR signaling, providing evidence for the existence of transmembrane intramolecular conformational changes in a single pass membrane protein. They also found that ligand binding induces intracellular conformational changes within monomeric EGFR, and implicate a series of electrostatic interactions in the conformational coupling. These results suggest that intramolecular transmembrane conformational changes upon ligand binding may be common to single-pass membrane proteins.

Scan and Unlock: A Programmable DNA Molecular Automaton for Cell-Selective Activation of Ligand-Based Signaling

The SUDA system uses embedded DNA-based chemical reaction networks to scan and evaluate molecular profiles of cell-surface proteins, achieving cell-specific signal modulation by quickly unlocking the protein-ligand in proximity to the target cell-surface. The versatility of the SUDA strategy is shown through engineering TNF alpha to induce programmed cell death of target cell subpopulations.

Single-molecule FRET imaging of GPCR dimers in living cells

Summary: This study presents a generally applicable method for using single-molecule fluorescence resonance energy transfer (smFRET) to detect and track transmembrane proteins diffusing within the plasma membrane of mammalian cells. The in-cell smFRET approach reveals agonist-induced structural dynamics within individual metabotropic glutamate receptor dimers. Evidence for receptor monomers, density-dependent dimers, and constitutive dimers in representative class A, B, and C receptors were observed using these methods.
