Extracellular SQSTM1 exacerbates acute pancreatitis by activating autophagy-dependent ferroptosis

Liangchun Yang et al.

Summary: This study reveals that extracellular SQSTM1 acts as a mediator of acute pancreatitis (AP) by enhancing sensitivity to autophagy-dependent ferroptotic cell death. High levels of serum SQSTM1 are found in AP patients and mice. Administration of SQSTM1-neutralizing antibodies protects mice against experimental AP.

Trophoblast-derived Lactic Acid Orchestrates Decidual Macrophage Differentiation via SRC/LDHA Signaling in Early Pregnancy

Lu Gao et al.

Summary: This study reveals the important role of lactic acid metabolism in the differentiation of decidual macrophages during early pregnancy, contributing to immune tolerance. Lactic acid can induce the polarization of M2 or M1 macrophages through regulation of oxidative phosphorylation and glycolysis under different oxygen levels. The inhibition of lactic acid metabolism through the HIF-1 alpha/SRC/LDHA pathway is involved in recurrent pregnancy loss. Blocking lactic acid intake rescues pregnancy in an abortion-prone mouse model, suggesting a potential therapeutic target in recurrent pregnancy loss.

Impairment of mitophagy and autophagy accompanies calcific aortic valve stenosis favouring cell death and the severity of disease

Giampaolo Morciano et al.

Summary: This observational study aims to establish the trend of autophagy in CAVS patients, analyze the involvement of other determinants impacting mitochondrial quality control mechanisms, and explore potential pharmacological interventions. The results show deficiencies in mitochondrial respiration, ATP production, and alterations in biogenesis and mitophagy pathways in the calcified valves, suggesting a rapamycin-based therapeutic strategy to revert pathological phenotypes. Boosting autophagy and mitophagy could potentially counteract cell death and sustain cell functions in the long term.
