1H detection and dynamic nuclear polarization-enhanced NMR of Aβ1-42 fibrils

Summary: Magic-angle spinning (MAS) NMR spectroscopy has been widely used to study the high-resolution structures of amyloid-beta (A beta) and other fibrils, but it suffers from low sensitivity. In this study, the drawbacks of MAS NMR were overcome by using two relatively recent techniques: H-1 detection and dynamic nuclear polarization (DNP). The results showed improved sensitivity and allowed for nearly complete resonance assignment and structural analysis of A beta fibrils.

Methyl-Driven Overhauser Dynamic Nuclear Polarization

Summary: The Overhauser effect, which requires the modulation of electron-nuclear hyperfine interactions, dominates dynamic nuclear polarization (DNP) in liquids and metals. This effect can also occur in insulating solids through rapid structural modulations linked to a modulation in hyperfine coupling. In this study, the addition of a methyl group to Blatter's radical was found to trigger Overhauser DNP. The rotation of the methyl group leads to a modulation of the hyperfine coupling to its protons, facilitating electron-nuclear cross-relaxation. The findings suggest the possibility of designing tailor-made Overhauser DNP polarizing agents for high-field MAS-DNP.

Organic Mixed-Valence Compounds and the Overhauser Effect in Insulating Solids

Summary: Recent experiments have demonstrated the ability of the BDPA radical to induce dynamic nuclear polarization in insulating solids, which was previously considered impossible. This unique capability is attributed to its mixed-valence nature and intramolecular charge transfer. Through advanced calculations and simulations, the class II mixed-valence nature of BDPA and the mechanism of electron hopping have been confirmed, paving the way for the development and fine tuning of novel polarizing agents.
