Deep structural insights into RNA-binding disordered protein regions

Summary: This article summarizes the current state of structural knowledge on the RNA binding modes of disordered protein regions and proposes a classification system based on their sequential and structural properties. Through detailed structural analysis of complexes containing disordered protein regions binding to RNA, two major binding modes representing different recognition strategies and functions were identified. Comparisons with DNA binding disordered proteins revealed key differences and similar binding strategies, suggesting a wider substrate acceptance by these proteins.

All-Atom Simulations Elucidate the Impact of U2AF2 Cancer- Associated Mutations on Pre-mRNA Recognition

Summary: In this study, we investigated the impact of U2AF2 cancer-associated mutations on polypyrimidine recognition through extensive molecular dynamics simulations and dynamical network analysis. Our results showed that these mutations affect the binding of U2AF2 to the polypyrimidine sequence, regardless of their location on the distinct domains of U2AF2. We also identified cross-communication pathways between the mutation sites and recognition hot spots, whose strength is altered by the mutations. These findings suggest a structural/dynamical interplay of U2AF2 domains underlying polypyrimidine recognition and reveal the impact of cancer-associated mutations on the cooperativity of U2AF2 domains in polypyrimidine selection.

Mechanism of RNA recognition by a Musashi RNA-binding protein

Summary: The Musashi RNA-binding proteins play important roles in regulating mRNA translation, cell stemness, and tumorigenesis. Musashi-1 (MSI1), previously known as an intestinal and neural stem cell marker, has been found to be overexpressed in many cancers and is a significant drug target for acute myeloid leukemia and solid tumors. In this study, researchers utilized all-atom microsecond-timescale simulations to investigate the dynamic mechanism of Numb RNA binding to MSI1. Their simulations revealed important intermediate conformational states during RNA binding and provide insight for structure-based drug design targeting MSI1 and other RBPs.
