Dissolution/Permeation of Albendazole in the Presence of Cyclodextrin and Bile Salts: A Mechanistic In Vitro Study into Factors Governing Oral Bioavailability

Jonas Borregaard Eriksen et al.

Summary: This study aimed to investigate the impact of the interaction between bile salts and cyclodextrins on the dissolution-permeation of poorly soluble drug compounds. The results showed that the interplay between bile salts and cyclodextrins can enhance the free fraction of the drug in solution to a greater extent than using either component alone.

Application of tiny-TIM as a mechanistic tool to investigate the in vitro performance of different itraconazole formulations under physiologically relevant conditions

Alvaro Lopez Marmol et al.

Summary: The increasing number of poorly water-soluble compounds in drug development presents a major challenge for oral drug delivery. This study utilized the tiny-TIM system, a realistic in vitro model of the human gastrointestinal tract, to investigate the performance of an itraconazole formulation under clinically relevant conditions. The results demonstrated the ability of this model to predict the effect of food intake and gastric pH modification on drug bioavailability.

Modulation of Paracellular-like Drug Transport across an Artificial Biomimetic Barrier by Osmotic Stress-Induced Liposome Shrinking

Jonas Borregaard Eriksen et al.

Summary: Treating the PermeaPad(R) barrier with low osmolality solutions appears to enhance the transport of hydrophilic compounds by inducing the osmotic shrinking of lipid vesicles, creating wider water channels for compound passage.

Using Tiny-TIM Dissolution and In Silico Simulation to Accelerate Oral Product Development of a BCS Class II Compound

Laibin Luo et al.

Summary: This study establishes in vitro-in vivo correlation and predicts oral bioavailability using in vitro data and in silico pharmacokinetic model. The predictive model has the potential to reduce animal and human experiments and expedite the drug development process, as it is consistent with clinical studies.
