HFIP-Assisted Single C-F Bond Activation of Trifluoromethyl Ketones using Visible-Light Photoredox Catalysis

Summary: This study presents a novel visible light photoredox catalytic method for the selective cleavage of a single strong C-F bond in trifluoromethyl ketones. The reaction shows excellent chemoselectivity and functional group tolerance under mild conditions, and high-level DFT calculations are used to elucidate the mechanism.

The role of hexafluoroisopropanol (HFIP) solvent in assisting the pTSA Bronsted acid catalyst and the current limitations of the carbonyl-olefin metathesis reaction were revealed through experimental and computational mechanistic studies.

Synthesis of salicylates from anionically activated aromatic trifluoromethyl group

Summary: This study describes an efficient approach to salicylates through a novel transformation of anionically activated aromatic trifluoromethyl group. The anionically activated trifluoromethyl group can react with phenols/alcohols under alkaline conditions to yield aryl/alkyl salicylates in high yields. Mechanistic studies have revealed that the carbonyl oxygen atom of the ester is derived from H2O in the solvent.

These new drugs are classified based on their chemical structures as biologics, TIDES, natural products, fluorine-containing molecules, nitrogen aromatic heterocycles, and other small molecules.

Summary: The unique properties of fluorine-containing organic compounds have led to the development of diverse C-F bond activation techniques, with a focus on pharmaceuticals and specialty materials. While advances have been made in functionalizing activated C-F bonds using transition metal complexes, fewer approaches are available for nonactivated C-F bonds due to the difficulty in oxidative addition of transition metals. Using Lewis acid and light/radical initiator to activate inert C-F bonds has emerged as a powerful tool, providing greener, more economical processes without heavy metal residues in the pharmaceutical industry.
