Insight into the Mechanism of the CuAAC Reaction by Capturing the Crucial Au4Cu4-π-Alkyne Intermediate

Yaping Fang et al.

Summary: A designed Au4Cu4/CNT nanocluster system catalyzes the CuAAC reaction of terminal alkynes without alkyne deprotonation, unlike the classic mechanism involving a sigma,pi-alkynyl intermediate. DFT results confirmed the activation of HC CPh by pi-complexation with Au4Cu4. The specificity of Au4Cu4 involving synergy between Cu and Au was shown by the inactivity of internal alkynes under the same conditions for both Au-11/CNT and Cu-11/CNT catalysts.

Summary: In this study, the structural, spectroscopic, and electronic properties of a newly synthesized compound were determined using experimental and theoretical methods. The results demonstrated that the applied level theory reproduced well the experimental results. Furthermore, molecular docking calculations indicated stable and favorable binding interactions, suggesting potential for synthesizing new coumarin derivatives with significant biological and pharmacological properties.
