By combining in vitro measured residence time, equilibrium measures of drug-target affinity, and in vivo measures of pharmacokinetic half-life, researchers can have a powerful approach to optimize drugs for clinical effect. Further studies are needed to better understand the dynamic interplay between residence time, target pathobiology, drug distribution, and drug pharmacokinetics despite the significant use and refinement of this model.
