Association of locus coeruleus integrity with Braak stage and neuropsychiatric symptom severity in Alzheimer's disease

Clifford M. Cassidy et al.

Summary: This study used neuromelanin-sensitive MRI (NM-MRI) to evaluate the correlation between locus coeruleus (LC) degeneration and the severity of Alzheimer's disease (AD) as well as neuropsychiatric symptoms (NPS). The results showed that decreased LC integrity was associated with AD severity, but preserving LC in AD patients may increase the risk of impulse control symptoms. NM-MRI holds promise as a practical biomarker for predicting NPS risk or guiding AD treatment.

Association between neuromelanin-sensitive MRI signal and psychomotor slowing in late-life depression

Kenneth Wengler et al.

Summary: This study showed that dopamine function abnormalities are related to slowed gait speed in older adults with late-life depression. While NM-MRI signal could predict treatment response to L-DOPA, it did not demonstrate associations with baseline gait speed, processing speed, or depression severity. Early findings suggest an increase in the NM-MRI signal after 3 weeks of L-DOPA treatment, indicating potential as a marker for dopamine-related psychomotor slowing.

Substantia nigra neuromelanin magnetic resonance imaging in patients with different subtypes of Parkinson disease

Lu Wang et al.

Summary: Neuromelanin (NM) concentration in patients with Parkinson's disease decreases gradually as the disease progresses, and differs significantly between different motor subtypes. Using NM-MRI technology can help evaluate the progression of Parkinson's disease and differentiate between tremor dominant and postural instability and gait difficulty subtypes.

Imaging the rapidly developing brain: Current challenges for MRI studies in the first five years of life

Ted K. Turesky et al.

Summary: Rapid changes in brain anatomy and physiology in the first five years of life pose challenges for developmental MRI studies. There is a trade-off between using age-appropriate methods for different developmental stages or using identical methods across stages, which can lead to biases. Addressing challenges in data acquisition, processing, and analysis is essential for optimizing developmental comparisons.

Neuromelanin-Sensitive Magnetic Resonance Imaging in Schizophrenia: A Meta-Analysis of Case-Control Studies

Lara Wieland et al.

Summary: The study found a significant elevation of contrast-to-noise ratio (CNR) in the substantia nigra in patients with schizophrenia, supporting neuromelanin as a potential biomarker for dopaminergic dysfunction. Further research is needed to evaluate this candidate marker in large, longitudinal cohorts and take into account potential effects of disease state, medication, and symptoms.

Cross-Scanner Harmonization of Neuromelanin-Sensitive MRI for Multisite Studies

Kenneth Wengler et al.

Summary: The study aimed to develop a method to harmonize NM-MRI data across different scanners and sites, using support vector machine and support vector regression to assess the impact of harmonization on biological variability. After harmonization, the classifier accuracy for scanners dropped to chance level, but there was no significant impact on age prediction results.

Imaging the Substantia Nigra in Parkinson Disease and Other Parkinsonian Syndromes

Yun Jung Bae et al.

Summary: Parkinson disease is characterized by loss of dopaminergic cells in the substantia nigra of the midbrain. Recent advances in MRI technology have enabled elucidation of underlying pathophysiological changes in the nigral structure, contributing to improved diagnosis and monitoring of the disease progression.
