Cellular senescence in the cholangiopathies

Pamela S. Bogert et al.

Summary: Cellular senescence is increasingly recognized as a pathologic feature in inflammatory liver diseases. This review provides an update on the interplay between cholangiocytes, cellular senescence, and the related diseases, and highlights the discovery of novel models, key pathways, and druggable targets for therapeutic interventions.

Ileal bile acid transporter inhibition in Cyp2c70 KO mice ameliorates cholestatic liver injury

Jennifer K. Truong et al.

Summary: This study found that inhibiting the ileal BA transporter can alleviate liver injury in Cyp2c70 KO mice by reducing hepatic inflammation, macrophage infiltration, and biliary cell proliferation. Additionally, reducing hepatic accumulation of bile acids can alleviate liver injury independent of the hydrophobicity of the bile acid pool.

Maralixibat for the treatment of PFIC: Long-term, IBAT inhibition in an open-label, Phase 2 study

Kathleen M. Loomes et al.

Summary: The study found that maralixibat has certain efficacy and safety in children with FIC1 or BSEP deficiencies. Some patients with nt-BSEP deficiency responded to maralixibat, showing significant reductions in pruritus and improvements in growth and quality of life. However, maralixibat had poor efficacy in patients with FIC1 deficiency or t-BSEP deficiency.

Cholangiopathy and Biliary Fibrosis in Cyp2c70-Deficient Mice Are Fully Reversed by Ursodeoxycholic Acid

Jan Freark de Boer et al.

Summary: The study found that Cyp2c70(-/-) mice, lacking MCAs, had different effects on microbiome composition and varied consequences between sexes. Male mice showed early high plasma BAs and transaminases, which decreased over time, while females experienced sustained elevated levels with age, impaired gut barrier function, and progression to bridging fibrosis. Treatments with UDCA normalized hepatic and intestinal functions in female Cyp2c70(-/-) mice, suggesting a role of BA hydrophobicity in disease development.

Microbial Hydroxysteroid Dehydrogenases: From Alpha to Omega

Heidi L. Doden et al.

Summary: Bile acids and glucocorticoids, important signaling molecules derived from cholesterol, undergo metabolism by hydroxysteroid dehydrogenases (HSDHs) encoded by both the host and gut microbiota. This biotransformation can alter the properties of the resulting products, affecting receptor activation, solubility, and toxicity. Microbial HSDHs play a role in diseases like colorectal cancer, liver cancer, prostate cancer, and polycystic ovary syndrome, and may have potential therapeutic applications as steroid pool modulators or drug targets.

Combined ASBT Inhibitor and FGF15 Treatment Improves Therapeutic Efficacy in Experimental Nonalcoholic Steatohepatitis

David J. Matye et al.

Summary: Combining ASBT inhibitor and FGF15 signaling activation improves therapeutic efficacy against NASH by reducing bile acid burden in the liver and limiting intestinal lipid absorption. The synergistic actions of the two treatments produce weight loss, reduce adipose inflammation, and attenuate hepatocellular organelle stress. Further clinical studies are needed to explore the potential of this combined treatment in humans.
