Lipoprotein size is a main determinant for the rate of hydrolysis by exogenous LPL in human plasma

Oleg Kovrov et al.

Summary: This study investigated the effects of natural variations in known LPL regulators on LPL activity and found that the key determinant for LPL activity was a lipid signature strongly correlated with the average size of VLDL particles, as well as apolipoprotein A5 levels.

Angiopoietin-like protein 4 (ANGPTL4) is an inhibitor of endothelial lipase (EL) while the ANGPTL4/8 complex has reduced EL-inhibitory activity

Yan Q. Chen et al.

Summary: In this study, it was found that levels of ANGPTL4/8 were significantly higher in type 2 diabetes patients compared to controls and were inversely correlated with HDL. Additionally, ANGPTL4 was shown to be a more potent inhibitor of EL than ANGPTL3.

Metabolic profiling of angiopoietin-like protein 3 and 4 inhibition: a drug-target Mendelian randomization analysis

Summary: The study found that genetic mimicry of ANGPTL4 inhibition and LPL enhancement have similar metabolic effects, while genetic mimicry of ANGPTL3 inhibition showed differing metabolic effects, suggesting potential involvement of pathways independent of LPL. Inhibition of ANGPTL4 and enhancement of LPL through genetic mimicry were associated with a lower risk of coronary heart disease and type 2 diabetes.

Regulation of plasma triglyceride partitioning by adipose-derived ANGPTL4 in mice

Kathryn M. Spitler et al.

Summary: Elevated plasma triglyceride levels are associated with metabolic disease, and angiopoietin-like protein 4 (ANGPTL4) plays a role in regulating these levels. Adipocyte-specific deficiency of ANGPTL4 in mice led to changes in triglyceride metabolism, including higher adipose LPL activity and increased triglyceride uptake. However, the beneficial effects on glucose homeostasis seen initially were lost after chronic high-fat feeding.

ANGPTL8 protein-truncating variant associated with lower serum triglycerides and risk of coronary disease

Pyry Helkkula et al.

Summary: Protein-truncating variants (PTVs) that impact dyslipidemia risk may indicate therapeutic targets for cardiometabolic disease. By studying the genetic associations between lipid levels and the risks of CAD or T2D, researchers identified two PTVs in ANGPTL4 and ANGPTL8 that were associated with lower disease risks and lipid levels in the Finnish population.

Angiopoietin-like 3 inhibition of endothelial lipase is not modulated by angiopoietin-like 8

Kelli L. Sylvers-Davie et al.

Summary: High plasma TG and low HDL-C levels are risk factors for atherosclerosis and cardiovascular disease, regulated in part by ANGPTL3. This study found that ANGPTL8 is not necessary for EL inhibition, indicating that targeting ANGPTL8 for hypertriglyceridemia or cardiovascular disease may have different outcomes than targeting ANGPTL3.
