The computations indicate that the QueG mechanism likely involves single-electron transfer from the Co(I)Cbl species to oQ.

The Biochemical Landscape of Riboswitch Ligands

Summary: More than 55 types of riboswitches that respond to small metabolites or elemental ions have been experimentally proven. These riboswitches primarily sense ligands that are fundamental and likely to have been relevant to ancient life forms, including the RNA World. This article discusses the ligands sensed by common riboswitches in modern bacteria and their implications for ancient biological processes. Despite representing most major aspects of metabolism, there are sensory gaps in key areas, which could reveal weaknesses in RNA's performance during RNA World evolution or identify opportunities for discovering new riboswitch classes.

Cobalamin Riboswitches Are Broadly Sensitive to Corrinoid Cofactors to Enable an Efficient Gene Regulatory Strategy

Summary: In bacteria, riboswitches are gene regulatory RNAs that directly bind and detect metabolites, controlling many essential metabolic processes. Cobalamin riboswitches are an exception as they can respond to multiple variants of cobalamin (vitamin B-12), known as corrinoids. This study found that cobalamin riboswitches vary in their specificity for corrinoids, with some being broadly responsive and others being more selective. Analyses and models suggest that cobalamin riboswitches indirectly differentiate among corrinoids by sensing differences in their structural conformation. These findings provide insights into how bacteria can sense and respond to the chemical diversity of essential metabolites.

Summary: This study found that cobinamide is a strong and multifaceted antioxidant that can neutralize various reactive oxygen species with fast reaction rates. Experimental results in cellular and model organisms showed that cobinamide outperformed other well-known antioxidants in reducing oxidative stress. In diabetic mice, cobinamide completely prevented pathological changes, including oxidative damage, DNA damage, and fibrosis in the heart. Therefore, cobinamide, as a promising new antioxidant, has potential applications in diseases with heightened oxidative stress.

It investigated the voltammetric behavior of Co(II) complexes with different azo dyes and developed sensitive and reproducible methods for cobalt determination with a detection limit of 10(-7) mol.L-1. The accuracy of the proposed method was confirmed by the method of standard additions and successfully applied for the determination of vitamin B12 in pharmaceutical formulations.

Mechanistic Studies on the Reaction between Aquacobalamin and the HNO Donor Piloty's Acid over a Wide pH Range in Aqueous Solution

Summary: The reaction between aquacobalamin/hydroxocobalamin and nitroxyl generated by Piloty's acid was studied over a wide pH range. It was found that in basic solutions, nitroxyl can react with hydroxocobalamin to form nitrosylcobalamin despite the inert nature of the compound. The rate-determining step in the reaction between hydroxocobalamin and nitroxyl changes depending on the concentration of Piloty's acid. Additionally, at neutral pH, rapid formation of CbINO was observed when Piloty's acid was stable and did not produce HNO.
