Age-dependent rise in IFN-γ competence undermines effective type 2 responses to nematode infection

Summary: This study reveals that spontaneous differentiation and expansion of Th1 cells with age restricts type 2 immune responses to intestinal nematode infections. Blocking early IFN-gamma and IL-12 signals decreases Th2/1 differentiation and enhances resistance.

MHC class I-independent activation of virtual memory CD8 T cells induced by chemotherapeutic agent-treated cancer cells

Summary: Chemotherapy-treated cancer cells can activate CD8 T cells in an MHC class I-independent manner, leading to the generation of virtual memory (VM) CD8 T cells which may provide superior therapeutic effects on MHC class I-deficient tumors.

Long-lived macrophage reprogramming drives spike protein-mediated inflammasome activation in COVID-19

Sebastian J. Theobald et al.

Summary: Our study reveals that the SARS-CoV-2 spike protein induces inflammasome formation and IL-1 beta release in macrophages from COVID-19 patients, but not in healthy individuals. Furthermore, macrophages isolated from convalescent COVID-19 patients show robust S-protein-driven inflammasome activation, indicating innate immune memory after recovery.

Virtual memory T cells orchestrate extralymphoid responses conducive to resident memory

Shiyue Hou et al.

Summary: This study reveals that virtual memory CD8(+) T cells rapidly infiltrate the lungs during a primary influenza infection, promoting early viral control. These cells consist of CCR2(+) and CCR2(-) subsets, which differentiate effector- and memory-poised functions directly in the tissue. The T-VM cells bridge innate reaction and adaptive memory by orchestrating an extralymphoid primary response in the infected tissue.

Cytomegalovirus mediates expansion of IL-15-responsive innate-memory cells with SIV killing function

Gema Mendez-Lagares et al.

Summary: Interindividual immune variability is influenced by environmental factors such as chronic infections like CMV. In young macaques, RhCMV infection led to significant changes in immune cell composition and function, including expansion of innate-memory CD8(+) T cells. Vaccination with RhCMV 68-1 vectors also resulted in expansion of functional CD8 cells capable of inhibiting SIV replication ex vivo, suggesting potential for other vaccines expressing IL-15 to achieve innate-memory expansion.
