Phenotype, Function, and Clinical Significance of CD26+ and CD161+ Tregs in Splenic Marginal Zone Lymphoma

Xinyi Tang et al.

Summary: This study found that regulatory T-cells (Treg) are more abundant in the spleens of patients with splenic marginal zone lymphoma (SMZL) compared to reactive spleens. These Tregs can be separated into CD161(+)Treg and CD26(+)Treg subsets. CD161(+)Tregs are increased in SMZL but have dysregulated immune function. CD161(+)Tregs are associated with a favorable prognosis in SMZL, while CD26(+)Tregs are associated with a poor prognosis. Activation of the IL2/STAT5 pathway contributes to the expansion of CD26(+)Tregs and may lead to a poor clinical outcome in SMZL.

CC chemokine receptor 2 (CCR2) expression promotes diffuse large B-Cell lymphoma survival and invasion

Quan-Quan Hu et al.

Summary: The study demonstrates that CCR2 overexpression is an independent prognostic marker for diffuse large B-cell lymphoma (DLBCL) and predicts overall survival and progression-free survival. Blockade of CCR2 signaling with a CCR2 antagonist inhibits tumor cell proliferation, migration, and anti-apoptosis ability by affecting the PI3K/Akt and p38 MAPK signaling pathways.

Follicular Lymphoma Microenvironment Characteristics Associated with Tumor Cell Mutations and MHC Class II Expression

Guangchun Han et al.

Summary: In this study, the tumor and immune cell populations of follicular lymphoma (FL) were characterized using single-cell RNA sequencing. Different subsets of T cells were identified, including cytotoxic CD4 T cells. Somatic mutations were found to be associated with reduced MHC expression on FL cells. The expression of MHCII genes by FL cells was correlated with significant differences in the proportions and immunophenotypic characteristics of T cells.

Recent updates on chimeric antigen receptor T cell therapy for hepatocellular carcinoma

Jiaojiao Guo et al.

Summary: CAR-T therapy is a novel tumor immunotherapy that has shown promising outcomes in leukemia and lymphoma by enabling T cells to specifically recognize tumor-associated antigens through genetic engineering. However, challenges remain in treating hepatocellular carcinoma, and overcoming factors like tumor antigen heterogeneity, immunosuppressive microenvironment, and adverse reactions are crucial for enhancing the efficacy of CAR-T cells in HCC patients.

Clinical and Biological Subtypes of B-cell Lymphoma Revealed by Microenvironmental Signatures

Summary: In this study, a transcriptomic analysis of the microenvironment of DLBCL was conducted, revealing four major lymphoma microenvironment categories associated with distinct biological aberrations and clinical behavior. This research also identified genetic and epigenetic mechanisms used by cancer cells to evade microenvironmental constraints and suggested the use of DNA hypomethylating agents in selected DLBCL patients. Additionally, new therapeutic vulnerabilities in the extracellular matrix composition were uncovered, providing potential targets for decreasing DLBCL proliferation in preclinical models.

Reduced CD5 on CD8+ T Cells in Tumors but Not Lymphoid Organs Is Associated With Increased Activation and Effector Function

Faizah Alotaibi et al.

Summary: CD5, a marker for T cells and a subset of B cells, is negatively correlated with anti-tumor activity in lung cancer patients, potentially impairing activation of anti-tumor T cells. T cells with high CD5 levels in tumors and lymphoid organs exhibit higher levels of activation and effector function, indicating that CD5 may be a therapeutic target for immunotherapeutic activation in cancer therapy.

The receptor of the colony-stimulating factor-1 (CSF-1R) is a novel prognostic factor and therapeutic target in follicular lymphoma

Juan Garcia Valero et al.

Summary: The study revealed that the three-way crosstalk of FL-FDC-macrophages in FL promotes tumor growth and M2-like protumoral phenotype through secretion of CCL2 and CSF-1. Elevated CSF-1 levels in grade 3A FL patients and high CSF-1R expression in FL biopsies were associated with reduced overall survival and risk of transformation. Inhibition of CSF-1R with pexidartinib disrupted FL-M2 positive crosstalk and showed antitumor effects by reducing M2-macrophage viability and polarization.
