Evaluation of the Herb-Drug Interaction Potential of Commonly Used Botanicals on the US Market with Regard to PXR- and AhR-Mediated Influences on CYP3A4 and CYP1A2

Summary: In this study, extracts of 30 popular botanicals used in herbal dietary supplements were tested for their effects on human pregnane X receptor (hPXR) and human aryl hydrocarbon receptor (hAhR) activity, as well as on the activity of drug metabolizing enzymes. The results showed that several botanicals could activate hPXR and hAhR, and induce the activity of drug metabolizing enzymes, which may pose risks for herb-drug interactions. Further clinical investigations are needed to determine the clinical relevance of these findings.
