Review: emerging drug therapies in inflammatory bowel disease

Laurie B. Grossberg et al.

Summary: This review examines the mechanisms of action and clinical trial data of emerging drug therapies for inflammatory bowel disease (IBD). The results show that several new therapies have demonstrated positive efficacy and safety data in early clinical trials for both ulcerative colitis (UC) and Crohn's disease (CD). As more treatments for IBD are approved, further research is needed to identify predictors of response and conduct head-to-head trials to guide therapeutic options for patients.

Bacteroides thetaiotaomicron relieves colon inflammation by activating aryl hydrocarbon receptor and modulating CD4+T cell homeostasis

Keying Li et al.

Summary: Inflammatory bowel disease (IBD) is associated with gut microbiome dysbiosis, and modulating the composition of the intestinal flora, such as through Bacteroides thetaiotaomicron, can alleviate colonic inflammation by influencing tryptophan metabolism and T cell subsets. The treatment with B. thetaiotaomicron promotes antiinflammatory Treg/Th2 cell differentiation while suppressing pro-inflammatory Th1/Th17 cells, ultimately decreasing colon inflammation. Additionally, B. thetaiotaomicron increases levels of aryl hydrocarbon receptor (AHR) ligands, leading to changes in transcription factor expression profiles within T cells and suggesting therapeutic potential for IBD treatment.

An Insight into the Roles of Dietary Tryptophan and Its Metabolites in Intestinal Inflammation and Inflammatory Bowel Disease

Xiaolan Li et al.

Summary: Inflammatory bowel disease is a complex gastrointestinal disorder closely related to individual's immune regulation, gene-microbiota interactions, and dietary factors. Tryptophan and its metabolic pathways play vital roles in regulating intestinal inflammation by influencing cytokines, immune cells, and intestinal microbial composition.

Berberine improves colitis by triggering AhR activation by microbial tryptophan catabolites

Wanghui Jing et al.

Summary: The study showed that berberine can effectively improve the pathological phenotype, gut barrier disruption, and colon inflammation in rats with DSS-induced colitis. Furthermore, berberine can alleviate gut dysbiosis in rats and improve gut barrier function through the regulation of microbial metabolites to activate AhR.

Medicinal plants from the 14th edition of the Russian Pharmacopoeia, recent updates

Alexander N. Shikov et al.

Summary: Herbal medicine in Russia has a long history, with the evolution of medicinal plant handling from handwritten herbalist manuscripts to Pharmacopoeias carefully traced, including discussions on the pharmacological effects, safety, and clinical data of traditional and officinal herbal plants. The 14th edition of the Russian Pharmacopoeia contains descriptions of 107 medicinal plants, with 25 new monographs added in comparison to the previous edition.

Impact of Bacterial Metabolites on Gut Barrier Function and Host Immunity: A Focus on Bacterial Metabolism and Its Relevance for Intestinal Inflammation

Naschla Gasaly et al.

Summary: The diverse and dynamic microbial community of the human gastrointestinal tract plays a vital role in supporting the development and function of the gut immune barrier. Microbiota-derived metabolites are crucial mediators of host-microbial interactions and can regulate inflammation and immune responses.

Akkermansia muciniphila and its outer protein Amuc_1100 regulates tryptophan metabolism in colitis

Zhenyang Gu et al.

Summary: The study suggests that the probiotic Akkermansia muciniphila (Akk) may alleviate ulcerative colitis (UC) by regulating tryptophan (Trp) metabolism, suppressing the kynurenine pathway (KP) independently of colon tissue, and promoting Trp metabolism enrichment mediated by fecal microbiota.
