Effect of prepartum source and amount of vitamin D supplementation on lactation performance of dairy cows

Summary: The objectives of this experiment were to determine the effects of supplementing 25-hydroxyvitamin D3 compared with vitamin D3 at 1 or 3 mg/d in late gestation on production outcomes of dairy cows. The results showed that feeding 25-hydroxyvitamin D3 increased colostrum and milk yield compared to vitamin D3. The effect of 25-hydroxyvitamin D3 on milk yield may be related to serum calcium concentrations.

Structural basis of TRPV5 regulation by physiological and pathophysiological modulators

Summary: Transient receptor potential vanilloid 5 (TRPV5) is a calcium-selective ion channel that plays a crucial role in maintaining calcium homeostasis in the kidney. This study investigates the regulation of TRPV5 by phosphatidylinositol 4,5-bisphosphate (PI(4,5)P-2) and calmodulin (CaM), as well as the effects of parathyroid hormone (PTH), metabolic acidosis, and protein kinase A (PKA) on TRPV5 activity. The findings reveal that low pH inhibits TRPV5 by preventing PI(4,5)P-2 activation, and PKA controls TRPV5 activity by preventing CaM binding and channel inactivation.

Effect of duration of exposure to diets differing in dietary cation-anion difference on Ca metabolism after a parathyroid hormone challenge in dairy cows

Summary: The experiment aimed to investigate the effects of exposure to an acidogenic diet on acid-base balance, mineral digestion, and response to PTH in dairy cows. Results showed that cows fed the acidogenic diet exhibited metabolic acidosis within 24 hours, with increased concentration of iCa in blood by day 3. Additionally, during PTH challenges, cows on the acidogenic diet had higher iCa concentration and AUC than those on a normal diet.

1,25(OH)2 vitamin D3 stimulates active phosphate transport but not paracellular phosphate absorption in mouse intestine

Summary: Intestinal absorption of phosphate is primarily mediated by the active transcellular pathway involving NaPi-IIb, stimulated by 1,25(OH)(2) vitamin D-3. The effects of hormonal regulation on other targets besides NaPi-IIb remain unknown. Administration of 1,25(OH)(2) vitamin D-3 in mice resulted in increased active transport of phosphate specifically in the jejunum, with no effect on the paracellular pathway. Both wild-type and Slc34a2-deficient mice showed similar responses to 1,25(OH)(2) vitamin D-3 in terms of hyperphosphaturia and changes in FGF23 and PTH levels.

Summary: The study aimed to investigate the effects of different levels of dietary cation-anion difference (DCAD) on acid-base balance, mineral metabolism, and health responses in pregnant nulliparous cows. Results showed that reducing DCAD led to decreased blood and urine pH, induced compensated metabolic acidosis prepartum, increased mineral absorption, but had no effect on phosphorus digestibility.
