Vascular burden and cognition: Mediating roles of neurodegeneration and amyloid PET

Julie Ottoy et al.

Summary: This study investigated the association between cerebrovascular burden and cognitive function, and found that the relationship was mainly mediated by localized neurodegeneration rather than amyloid beta deposition. The results suggest an amyloid-independent pathway through which vascular burden affects cognitive function.

18F-florbetapir PET as a marker of myelin integrity across the Alzheimer's disease spectrum

Alexis Moscoso et al.

Summary: Recent evidence suggests that PET imaging with amyloid-beta tracers can be used to assess myelin integrity in cerebral white matter. The study found that demyelination levels progress across the AD continuum and are associated with clinical progression, indicating a relevant degenerative feature in the disease course.

Comparison of 11C-Pittsburgh Compound B and 18F-Flutemetamol White Matter Binding in PET

Burcu Zeydan et al.

Summary: This study investigates the use of PET imaging techniques for assessing white matter integrity and demyelination. The results show that both 11C-PiB and 18F-flutemetamol can effectively differentiate between white matter hyperintensities and normal-appearing white matter, and their uptake patterns are associated with age.

Amyloid PET imaging and dementias: potential applications in detecting and quantifying early white matter damage

Anna M. Pietroboni et al.

Summary: This study evaluates the use of amy-PET in patients with Alzheimer's disease and non-AD dementia. The results suggest that amy-PET is useful for assessing microstructural white matter injury in non-AD dementia, but may not be suitable for evaluating white matter damage in AD patients due to amyloid accumulation.

Venous Collagenosis as Pathogenesis of White Matter Hyperintensity

David Lahna et al.

Summary: The study indicates that venous collagenosis is associated with periventricular white matter hyperintensities (pvWMHs), while arterial collagenosis is not. This suggests the importance of changes in the venous system in the commonly observed MRI white matter lesions in older individuals.

In vivo myelin imaging and tissue microstructure in white matter hyperintensities and perilesional white matter

Jennifer K. Ferris et al.

Summary: White matter hyperintensities have negative effects on white matter structure and cognitive decline. Diffusion tensor imaging detects changes in white matter microstructure, while myelin water imaging provides specific markers of myelin content and interstitial fluid. This study found a spatial gradient of white matter metrics within the hyperintensity and perilesional white matter. Normal-appearing white matter adjacent to hyperintensities exhibits characteristics of a transitional stage.

Improved Segmentation of the Intracranial and Ventricular Volumes in Populations with Cerebrovascular Lesions and Atrophy Using 3D CNNs

Emmanuel E. Ntiri et al.

Summary: The study introduces two algorithms, iCVMapper and VentMapper, utilizing a convolutional neural network to segment ICV and ventricles from MRI data. Trained on a large dataset of older adults, the models outperformed other methods in evaluation metrics, demonstrated high time efficiency, and robustness to images with lower resolution and signal-to-noise ratio.

Free-water diffusion tensor imaging improves the accuracy and sensitivity of white matter analysis in Alzheimer's disease

Maurizio Bergamino et al.

Summary: Standard DTI and FW-DTI both showed decreased FA in AD, but higher AxD and RD were observed with standard DTI. The FW index was significantly elevated in AD, emphasizing the impact of free water on standard DTI in neurodegenerative pathologies. FW-DTI provided improved consistency in DTI metrics related to WM integrity in Alzheimer's disease.
