The pyroptosis-related gene signature predicts prognosis and indicates immune activity in hepatocellular carcinoma

Min Deng et al.

Summary: This study investigates the expression levels of pyroptosis-related genes in hepatocellular carcinoma (HCC) and their relationship with prognosis. A 10-gene signature was constructed to classify HCC patients into low-risk and high-risk groups, and the results showed that the low-risk group had significantly better survival rates. PRGs play a significant role in tumor immunity and have the potential to predict the prognosis of HCC patients.

Systemic Therapy for Hepatocellular Carcinoma: Chinese Consensus-Based Interdisciplinary Expert Statements

Yongkun Sun et al.

Summary: Hepatocellular carcinoma (HCC) is a common type of liver cancer that causes many cancer-related deaths worldwide. Though a molecular targeted drug has been available for the treatment of advanced HCC, no Chinese guidelines for novel molecular targeted therapies have been established. An interdisciplinary panel has reviewed the treatment of advanced HCC and provided an overview of molecular targeted therapies for healthcare professionals.

Could We Predict the Response of Immune Checkpoint Inhibitor Treatment in Hepatocellular Carcinoma?

Choong-kun Lee et al.

Summary: The use of anti-PD-1/PD-L1 is an important strategy for treating HCC, but its efficacy is limited to a small percentage of patients. The selection of patients for this treatment is crucial, and there is a need for biomarkers to predict the response. This review explores clinical and translational biomarkers for anti-PD-1/PD-L1 treatment in HCC, including etiological factors, disease extent, and inflammatory indices.

Laminin gamma 2-mediating T cell exclusion attenuates response to anti-PD-1 therapy

Lei Li et al.

Summary: PD-1/PD-L1 blockade therapies have shown significant clinical benefits for advanced cancer patients, but the effectiveness of treatment can be hindered by the up-regulation of laminin gamma 2 (Ln-gamma 2) in the tumor microenvironment, which is transcriptionally activated by transforming growth factor-beta 1 (TGF-beta 1) through JNK/AP1 signaling. Co-administration of a TGF-beta receptor inhibitor and chemotherapy drugs can enhance the anti-tumor activity of anti-PD-1 therapy.

Ferroptosis-related gene signature predicts prognosis and immunotherapy in glioma

Rong-Jun Wan et al.

Summary: A majority of ferroptosis-related genes (FRGs) showed differential expression among GBM, LGG, and NON-TUMOR groups (96.6%). Glioma patients with low-risk score had a more favorable clinical outcome, regardless of their tumor grade. Functional analysis revealed a positive correlation between high-risk score group and immune checkpoint blockade-related positive signatures, highlighting the critical role of glioma immunotherapy based on the risk score.

Tumor Immune Microenvironment and Immunosuppressive Therapy in Hepatocellular Carcinoma: A Review

Kyoko Oura et al.

Summary: Liver cancer, specifically hepatocellular carcinoma (HCC), has the fourth highest mortality rate among all cancers worldwide. Despite advancements in systemic therapy, HCC still has a poor prognosis due to drug resistance, recurrence, and metastasis. Recent therapeutic strategies, such as cancer immunosuppressive therapy utilizing immune checkpoint inhibitors (ICIs) and VEGF inhibitors, have shown promise in extending the lives of patients with advanced HCC.

A Focal Adhesion-Related Gene Signature Predicts Prognosis in Glioma and Correlates With Radiation Response and Immune Microenvironment

Haonan Li et al.

Summary: This study identified a differential gene expression signature related to glioma using bioinformatic databases, constructing an eight-gene FADG signature. Results showed significantly worse prognosis in the high-risk group, with risk scores positively correlated with genes related to DNA repair. The signature can predict prognosis in patients receiving radiation therapy and is associated with tumor microenvironment, immune cell infiltration, and immune checkpoint expression.
