Microbial production of vitamin B5: current status and prospects

In this study, a deep learning approach (DLKcat) was developed to predict k(cat) values for metabolic enzymes using only substrate structures and protein sequences. The predicted k(cat) values showed good performance in a genome-scale analysis of more than 300 yeast species, and were able to explain phenotypic differences.

Strengthening the (R)-pantoate pathway to produce D-pantothenic acid based on systematic metabolic analysis

Summary: Through systematic metabolic engineering, higher yielding D-pantothenic acid producing strains were obtained based on previously developed strains. Overexpression of lpd gene, disruption of non-PTS pathway, and enhancement of PTS pathway were conducted to increase precursor accumulation. Blocking pyruvate bypass pathway, enhancing (R)-pantothenic acid pathway, introducing heterologous proteins, and blocking other synthesis pathways significantly increased the yield of D-pantothenic acid to 6.33 g/L.

The optimized conditions led to increased D-PA titer in shake flask fermentation and fed-batch fermentation, with significant reduction in acetate accumulation. These findings highlight the importance of medium composition and cell growth rate in increasing D-PA concentration for microbial fermentation.
