Eco-friendly sequential one-pot synthesis, molecular docking, and anticancer evaluation of arylidene-hydrazinyl-thiazole derivatives as CDK2 inhibitors

Abeer M. El-Naggar et al.

Summary: This study describes the discovery and development process of a novel potential CDK2 inhibitor. The synthesized compounds showed strong growth inhibition against various cancer cell lines with low toxicity on normal fibroblast cells. One compound exhibited inhibitory effects on cell cycle progression and apoptosis. Molecular docking study identified the structural elements involved in the inhibitory activity and rationalized the structure-activity relationship.

Summary: This study involved the design, synthesis, and evaluation of urea/thiourea derivatives containing heteroaromatic scaffolds, indicating activity against gram-positive bacteria. Molecular docking studies revealed interactions with proteins involved in peptidoglycan synthesis. ADME calculations suggested oral absorption for these compounds.

Synthesis and antiproliferative screening of novel doubly modified colchicines containing urea, thiourea and guanidine moieties

Julia Krzywik et al.

Summary: A new series of 10-demethoxy-10-methylaminocolchicines with urea, thiourea or guanidine moieties at position C7 have been designed, synthesized and evaluated for in vitro anticancer activity. Most of the new derivatives showed promising anticancer activity in the nanomolar range, with lower IC50 values compared to cisplatin or doxorubicin. Specifically, two ureas and two thioureas displayed good antiproliferative effects, making them potential candidates for further research in the field of anticancer drugs based on the colchicine skeleton.
