Erythrocyte transglutaminase-2 combats hypoxia and chronic kidney disease by promoting oxygen delivery and carnitine homeostasis

Ping Xu et al.

Summary: This study reveals the crucial role of erythrocyte transglutaminase-2 (eTG2) in triggering oxygen delivery and tissue metabolic reprogramming. eTG2-mediated post-translational modification is essential for promoting oxygen delivery in adapting to high altitude and hypoxia. In a model of chronic kidney disease, eTG2 is critical for maintaining carnitine homeostasis and combating renal hypoxia-induced changes. Carnitine supplementation is identified as an effective and safe therapeutic approach to counteract hypertension and CKD progression.

Donor sex, age and ethnicity impact stored red blood cell antioxidant metabolism through mechanisms in part explained by glucose 6-phosphate dehydrogenase levels and activity

Angelo D. Alessandro et al.

Summary: Storage of red blood cells in the blood bank leads to metabolic alterations that affect their ability to resist oxidative stress, with donor characteristics such as sex, age, ethnicity, additive solution type, and glucose-6-phosphate dehydrogenase status influencing the metabolism and susceptibility to hemolysis of stored erythrocytes following oxidative insults. Metabolomic markers of hemolysis and glucose-6-phosphate dehydrogenase deficiency were identified, revealing dysregulation in detoxification pathways of oxidized lipids in extreme oxidative hemolysis.

Protein-L-isoaspartate O-methyltransferase is required for in vivo control of oxidative damage in red blood cells

Angelo D'Alessandro et al.

Summary: Red blood cells face the challenge of high levels of reactive oxygen species and lack the ability to synthesize new gene products. While progress has been made in understanding how RBC neutralize reactive oxygen species, little is known about how they repair damage caused by oxidative stress. RBC can only repair oxidized proteins directly and face challenges in repairing damage caused by reactive oxygen species.

Chaperone-mediated autophagy sustains haematopoietic stem-cell function

Shuxian Dong et al.

Summary: Activation of hematopoietic stem cells involves molecular adaptations, with Chaperone-Mediated Autophagy (CMA) playing a role in sustaining HSC function and declining with age. Restoring old HSC functionality through CMA activation suggests it may be a promising therapeutic target for conditions like aging or stem-cell transplantation.

Rapid clearance of storage-induced microerythrocytes alters transfusion recovery

Camille Roussel et al.

Summary: This study found that a high proportion of SMEs in RBC concentrates from healthy volunteers correlated with poor transfusion recovery. When perfused through a human spleen, 15% and 61% of long-stored RBCs and SMEs were cleared within 70 minutes, respectively. High initial proportion of SMEs also correlated with high retention of RBCs by perfused human spleen.
