Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Yuanhuacin and Related Anti-Inflammatory and Anticancer Daphnane Diterpenes from Genkwa Flos-An Overview

Christian Bailly

Summary: The dried flower buds of Daphne genkwa Sieb. et Zucc. have long been used in traditional Chinese medicine to treat inflammatory diseases. Various diterpenoids, including the yuanhuacin (YC) series and analogues, have been isolated from Genkwa Flos. These compounds have shown potential as anticancer agents, with YC being particularly effective against lung and breast cancers. Although protein kinase C has been identified as a key factor in YC's activity, the exact molecular targets of these compounds remain unclear. The pharmacological activity of each compound and the properties of Genkwa Flos extracts are discussed, along with the potential toxic effects associated with their use.

Targeting the p53-MDM2 pathway for neuroblastoma therapy: Rays of hope

Atif Zafar et al.

Summary: Neuroblastoma poses a significant challenge in pediatric oncology, with p53 protein playing a key protective role against genome instability. Overexpression of MDM2 in neuroblastoma can lead to p53 inhibition, drug resistance, and other non-canonical p53 functions. Research is focused on restoring p53 function by targeting the p53-MDM2 axis for potential therapeutic strategies.

Targeting metastatic cancer

Karuna Ganesh et al.

Summary: Improved understanding of metastases biology has led to advancements in targeting and treating both micro- and macrometastases. Recent studies have identified the unique biology of metastasis-initiating cells, providing new opportunities for developing more effective treatment strategies through cancer sequencing, mechanistic studies, and clinical trials. These insights offer potential for better outcomes in targeting metastatic relapse and improving patient survival.

Up-Regulation of LINC00665 Facilitates the Malignant Progression of Prostate Cancer by Epigenetically Silencing KLF2 Through EZH2 and LSD1

Peng Xue et al.

Summary: This study indicates that LINC00665 is significantly up-regulated in prostate cancer tissues and cells, while knock-down of LINC00665 significantly attenuates the proliferation and migration ability of PCa cells. Mechanically, LINC00665 epigenetically silences KLF2 expression by binding to EZH2 and LSD1, thereby promoting the malignant progression of PCa.

The Most Competent Plant-Derived Natural Products for Targeting Apoptosis in Cancer Therapy

Sadegh Rajabi et al.

Summary: Cancer is a challenging global health problem, and understanding the six cancer hallmarks is crucial in developing novel therapies. Apoptosis, involving intrinsic and extrinsic signaling pathways, plays a critical role in cancer treatment, and targeting multiple molecules in these pathways may offer new treatment options and prevent resistance. Plant extracts and plant-derived molecules have shown potential in activating apoptosis pathways in human cancer cells, making them promising candidates for cancer treatment.

Yuanhuacine Is a Potent and Selective Inhibitor of the Basal-Like 2 Subtype of Triple Negative Breast Cancer with Immunogenic Potential

Charles S. Fermaintt et al.

Summary: The study identified a class of plant-derived natural products, including yuanhuacine, that selectively kill cells representing a molecularly defined subtype of triple negative breast cancer (TNBC) and promote an antitumor immune response. Yuanhuacine exerts potent antitumor efficacy in a mouse TNBC model by activating protein kinase C.

Cancer incidence, mortality, and burden in China: a time-trend analysis and comparison with the United States and United Kingdom based on the global epidemiological data released in 2020

Haibo Qiu et al.

Summary: Cancer is a major cause of death and economic burden in China, with lower incidence but higher mortality and DALY rates compared to the US and UK. The cancer spectrum in China is changing, with increasing burden of certain cancers and a shift towards patterns seen in developed countries.
