Multiplexed electrospray enables high throughput production of cGAMP microparticles to serve as an adjuvant for a broadly acting influenza vaccine

Summary: This study reports a new method of encapsulating an interferon gene stimulator using electrospray, which significantly improves its biological activity. A multiplexed electrospray apparatus with higher throughput than a single-head apparatus was also developed. In vitro and in vivo experiments demonstrated that microparticles produced using this high throughput process are an effective vaccine adjuvant to induce a balanced immune response.

Summary: Infection and vaccination expose individuals to influenza antigens, but the production of broadly reactive influenza antibodies is limited. This study found that inducing broadly reactive influenza antibodies can lead to an increase in autoreactive antibodies and exacerbate autoimmune diseases. This suggests that self-tolerance mechanisms play a role in limiting the presence of broadly reactive influenza antibodies, which can worsen disease in certain conditions.

This study presents a vaccine strategy using a scalable microparticle platform containing a STING agonist as an adjuvant, which provides durable protection against the influenza virus. Animal studies demonstrate that this vaccine-adjuvant combination can reduce viral shedding, improve vaccine outcomes, and provide sustained protection even a year after a single dose.
