Functionalized niosomes as a smart delivery device in cancer and fungal infection

Summary: Many diseases remain untreated due to lack of suitable therapeutic drugs or drug delivery devices. Niosomes, as drug delivery carriers, have advantages such as ease of surface modification, non-immunogenicity, and cost-effectiveness. Active targeting through coupling suitable ligands on niosomal surface is a preferred method for treatment. Combination therapy and gene silencing concept are also advanced alternative treatments for certain diseases.

Summary: WF-208, inspired by the anti-tubercular activity of isoniazid and 5-bromoisatin, demonstrated potent efficacy against MDR and XDR strains of M. tuberculosis. Incorporation of WF-208 into nanosized vesicles not only enhanced stability and sustained antimicrobial effects, but also significantly increased its anti-mycobacterial activity.

Formulation and Evaluation of Hybrid Niosomal In Situ Gel for Intravesical Co-Delivery of Curcumin and Gentamicin Sulfate

Summary: The current study describes the development of a hybrid drug delivery platform using thermosensitive niosomal in situ gels for intravesical drug delivery. The optimized formulation achieved high entrapment efficiency and suitable physicochemical properties. The simultaneous loading of two drugs in the gel resulted in a synergistic effect, leading to sustained release. Therefore, this thermosensitive niosomal in situ gel can be considered as a feasible platform for intravesical drug delivery.

Identification of novel furo[2,3-d]pyrimidine based chalcones as potent anti-breast cancer agents: synthesis, in vitro and in vivo biological evaluation

Summary: Synthetic furo[2,3-d]pyrimidine-based chalcones exhibited potent anti-proliferative and cytotoxic activities against cancer cells. Among the halogen-bearing derivatives, compounds 5d and 5e showed significant anti-proliferative activity against a resistant cell line and comparable efficacy to doxorubicin in in vivo anticancer assessment.

Novel oxindole/benzofuran hybrids as potential dual CDK2/GSK-3β inhibitors targeting breast cancer: design, synthesis, biological evaluation, and in silico studies

Summary: In this study, three series of oxindole-benzofuran hybrids were designed and synthesised as dual CDK2/GSK-3 beta inhibitors targeting breast cancer. The N-1 -unsubstituted oxindole derivatives showed potent cytotoxic activity, while the N-1-substituted derivatives exhibited moderate to weak activity on breast cancer cell lines. The most potent compounds caused cell cycle arrest in the G2/M phase by inhibiting CDK2/GSK-3 beta, as demonstrated by enzyme inhibition assays and molecular docking studies.

Summary: The study identified a redox homeostasis network involving the iron-sulfur cluster enzyme NFS1 in the survival mechanisms governed by the tumor hypoxia-induced pH regulator CAIX. Targeting CAIX while depleting NFS1 or blocking cyst(e)ine availability enhanced ferroptosis and inhibited tumor growth significantly. Altering intracellular pH and iron homeostasis through inhibiting CAIX activity may lead to the development of innovative therapeutic strategies for solid tumors to overcome hypoxia- and acidosis-mediated tumor progression and therapeutic resistance.
