Monitoring polysorbate hydrolysis in therapeutic proteins using an ultrasensitive extraction-free fatty acid quantitation method

Summary: A rapid lipase activity assay was introduced for accurate quantitation of free fatty acids without the need for a fatty acid extraction step. This assay can detect low levels of PS degradation within 1 day and the released free fatty acids correlate strongly with the degree of PS20 degradation.

Mechanistic understanding of metal-catalyzed oxidation of polysorbate 80 and monoclonal antibody in biotherapeutic formulations

Summary: This study evaluated the oxidation of polysorbate 80 (PS80) and a monoclonal antibody (mAb) under different conditions. The presence of iron accelerated the oxidation of PS80 and increased the oxidation of a specific solvent-exposed mAb methionine. PS80-AO was more sensitive to oxidative degradation than PS80-MC. The kinetics of PS80 oxidation was pH-dependent in the presence of histidine buffer. Increasing mAb concentration exhibited a protective effect against metal catalyzed PS80 and methionine oxidation.

A Mechanistic Understanding of Monoclonal Antibody Interfacial Protection by Hydrolytically Degraded Polysorbate 20 and 80 under IV Bag Conditions

Summary: The purpose of this study was to understand the impact of degradation products of polysorbates (PS20 and PS80) in liquid drug products on mAb1 aggregation. The study found that the degradation products were primarily composed of free fatty acids and POE head groups, and the influence of fatty acids on surface tension was greater than that of ester distribution. In the presence of laurate salts, mAb1 aggregation increased, potentially due to the increased self-association of mAb1 in the presence of laurate salts. These results help explain the differences in mAb1 aggregation between PS20 and PS80 under IV bag agitation conditions.

Surface Tension and Self-association Properties of Aqueous Polysorbate 20 HP and 80 HP Solutions: Insights into Protein Stabilisation Mechanisms

Summary: The study focused on determining the self-association properties of compendial grade polysorbate 20 HP (PS 20 HP) and polysorbate 80 HP (PS 80 HP) regarding micelle size, critical micelle concentration range (cmr), and surface tension. Results showed that PS 80 HP formed larger micelles compared to PS 20 HP, with critical micelle concentration range values (cmr) at 25 degrees Celsius being considerably lower for PS 80 HP. The temperature dependency of the micelle size was more pronounced for PS 80 HP.

High-Throughput, Fluorescence-Based Esterase Activity Assay for Assessing Polysorbate Degradation Risk during Biopharmaceutical Development

The assay allows for rapid characterization of protein samples in under three hours, with the esterase activity directly correlating with polysorbate degradation, providing valuable information on degradation risk throughout drug development.
