Mechanisms of nanoparticle radiosensitization

Ivan Kempson

Summary: Metal-based nanoparticles have shown promise in enhancing the effects of radiotherapy, with potential for significant improvements in outcomes. However, the diverse mechanisms and variability between different nanoparticles highlight the need for individualized research and optimization. Similar to anti-cancer drugs, nanoparticle formulations may need to be tailored for specific indications to achieve the best therapeutic outcomes.

Theranostic AGuIX nanoparticles as radiosensitizer: A phase I, dose-escalation study in patients with multiple brain metastases (NANO-RAD trial)

Camille Verry et al.

Summary: The study assessed the safety and feasibility of combining AGuIX nanoparticles with whole brain radiotherapy for patients with brain metastases, showing no dose-limiting toxic effects and clinical benefits for most patients. Efficient targeting of metastases and correlation between contrast enhancement and tumor response were observed, supporting a potential radiosensitizing effect. Ongoing phase II studies will further evaluate efficacy.
