Itaconate and fumarate derivatives inhibit priming and activation of the canonical NLRP3 inflammasome in macrophages

Christopher Hoyle et al.

Summary: This study found that metabolite derivatives of itaconate and fumarate can inhibit the activation of NLRP3 inflammasome and reduce the secretion of inflammatory cytokines, suggesting their potential therapeutic role in diseases such as multiple sclerosis.

Preclinical Evaluation of Dimethyl Itaconate Against Hepatocellular Carcinoma via Activation of the e/iNOS-Mediated NF-κB-Dependent Apoptotic Pathway

Anurag Kumar Gautam et al.

Summary: In this study, it was found that dimethyl itaconate (DI) can effectively inhibit the development of hepatocellular carcinoma (HCC) in vivo by regulating multiple signaling pathways and molecular mechanisms. Through the modulation of mitochondrial apoptosis, the NF-kappa B pathway, and metabolomic profiles, DI shows promising anti-HCC activity in DEN-induced HCC rats.

Characterization of the activity and the mechanism of action of a new toluquinol derivative with improved potential as an antiangiogenic drug

Jose Antonio Torres-Vargas et al.

Summary: Inhibition of sustained angiogenesis is an effective approach for treating cancer, blindness, and other diseases. This study explored the structural modifications of a natural compound called toluquinol to develop improved drug candidates with antiangiogenic activity. The results showed that some derivatives of toluquinol could maintain or even improve the antiangiogenic activity while exhibiting lower cytotoxicity. The mechanism of action may involve apoptosis induction, increased reactive oxygen species levels, decreased redox capacity, and inhibition of VEGFR2, Akt, and ERK phosphorylation. The newly developed angiogenesis inhibitor shows promising potential for treating angiogenesis-related diseases due to its biological activity and lower undesired toxicity.

The role of itaconate in host defense and inflammation

Christian G. Peace et al.

Summary: Inflammatory stimuli cause metabolic reprogramming in macrophages, particularly in the Krebs cycle, resulting in the accumulation of itaconate and other metabolites. Itaconate plays diverse roles in immunity and inflammation, including inhibiting inflammasome activation and glycolysis, activating anti-inflammatory transcription factors, and exerting anti-inflammatory effects in various disease models.

Dimethyl Itaconate Alleviates the Inflammatory Responses of Macrophages in Sepsis

Sheng Zhang et al.

Summary: Dimethyl itaconate exhibits anti-inflammatory effects in sepsis, increasing survival rate, reducing levels of inflammatory cytokines, and ameliorating lung injury. It functions by activating Nrf2 to inhibit cytokine production.

Dimethyl itaconate inhibits LPS-induced microglia inflammation and inflammasome-mediated pyroptosis via inducing autophagy and regulating the Nrf-2/HO-1 signaling pathway

Su Yang et al.

Summary: The study revealed that DI inhibited NLRP3 assembly, LDH release, and GSDMD cleavage. Co-treatment with DI, LPS, and ATP promoted the transition from M1 to M2, reduced inflammatory mediator expression, and inhibited NF-kappa B phosphorylation. Additionally, DI reduced reactive oxygen species production through the Nrf-2/HO-1 pathway and induced cellular autophagy, playing a key role in microglia inflammation and NLRP3 inflammasome-mediated pyroptosis.
