Single-cell transcriptomic characterization of a gastrulating human embryo

Richard C. Tyser et al.

Summary: The single-cell transcriptional profile of a human embryo between 16 and 19 days after fertilization shows similarities and differences in gastrulation compared to mouse and non-human primate models. This study provides new insights into human development and offers valuable information for directed differentiation of human cells in vitro.

Modulating mesendoderm competence during human germ layer differentiation

James R. Valcourt et al.

Summary: As pluripotent human embryonic stem cells differentiate towards a specific germ layer, they no longer have the ability to adopt alternative fates. Research shows that as cells progress along the ectoderm trajectory, their probability of adopting a mesendodermal fate decreases sharply. By studying the gene regulatory network and key transcription factors, researchers can modulate the window of competence to control the fate choices of stem cells during differentiation.
