Integrase Inhibitor Resistance Mechanisms and Structural Characteristics in Antiretroviral Therapy-Experienced, Integrase Inhibitor-Naive Adults with HIV-1 Infection Treated with Dolutegravir plus Two Nucleoside Reverse Transcriptase Inhibitors in the DAWNING Study

Mark Underwood et al.

Summary: In the phase IIIb DAWNING study, treatment-experienced adults who received dolutegravir showed emergence of treatment-emergent INSTI resistance-associated substitutions, with some participants developing mutations like G118R and R263K. These mutations were associated with a significant increase in dolutegravir resistance compared to baseline levels. Overall, the study highlights the complex pathway to dolutegravir resistance involving HIV-1 phenotypic changes and reduced viral fitness.

T helper type (Th1/Th2) responses to SARS-CoV-2 and influenza A (H1N1) virus: From cytokines produced to immune responses

Elahe Aleebrahim-Dehkordi et al.

Summary: Cytokines produced by T helper cells play crucial roles in host defense against viruses, with Th1 cells essential for defense against intracellular pathogens and Th2 cells critical for parasite elimination. Diseases caused by SARS-CoV-2 and H1N1 influenza have led to global epidemics, with pulmonary immunopathology being a leading cause of mortality. Inadequate or excessive levels of specific cytokines can have adverse effects on the body during these viral infections.

Summary: This study analyzed the available NA protein sequences of H1N1 influenza viruses worldwide and found that 3.76% of H1N1 viruses currently harbor oseltamivir resistance, with the most common mutation being H274Y. Oseltamivir resistance is mainly found in humans, swine, and avian, and the resistance level peaked in 2007-2008 and quickly decreased in 2009. The study highlights the importance of globally monitoring oseltamivir resistance.

Alloferon Affects the Chemosensitivity of Pancreatic Cancer by Regulating the Expression of SLC6A14

Hyejung Jo et al.

Summary: Pancreatic cancer is a highly lethal solid tumor with limited treatment options. Recent studies have shown that upregulation of the amino acid transporter SLC6A14 is associated with glutamine metabolism in some cancers. Alloferon, a peptide derived from the insect immune system, has been found to have anti-tumoral effects, although its underlying mechanisms are not well understood. In this study, researchers investigated the effects of alloferon on pancreatic cancer cell lines and found that it downregulated SLC6A14 expression and reduced glutamine uptake. This suggests that alloferon could potentially be used as an adjuvant for chemotherapeutic drugs in pancreatic cancer treatment.

Oseltamivir modified bovine serum albumin inhibits neuraminidase activity and accumulates virion particles to disturb influenza virus replication

Hai-Juan Qin et al.

Summary: The preparation of an OS-BSA conjugate for use as a multivalent NA inhibitor is reported, showing superior inhibitory effects compared to the monomeric OS azidohexyl ester. Further investigation revealed that the prepared OS-BSA has a dual role as a multivalent NA inhibitor and an adsorbent for virion particles, contributing to the inhibition of influenza viral replication. This study provides insight into the antiviral mechanism of multivalent NA inhibitors and lays the foundation for the development of novel antiviral agents.
