Separation of phosphorothioated oligonucleotide diastereomers using multiplexed drift tube ion mobility mass spectrometry

Summary: Hydrophilic interaction liquid chromatography coupled to drift tube ion mobility spectrometry was used to separate diastereomers of five-unit oligonucleotides with different phosphorothioate linkages. The study investigated multiplexed drift tube ion mobility and post-acquisition processing using a demultiplexing tool. By optimizing the electric field inside the drift tube, the diastereomers could be separated unambiguously. The use of multiplexed ion mobility and a high-resolution demultiplexing tool represents a breakthrough in resolving diastereomers in linear drift tube ion mobility spectrometry.

Pharmacokinetics and pharmacodynamics of inclisiran, a small interfering RNA therapy, in patients with hepatic impairment

Summary: This study investigated the pharmacokinetics, pharmacodynamics, and safety of Inclisiran in patients with mild or moderate hepatic impairment compared to those with normal hepatic function. The results showed that while there was an increase in pharmacokinetic exposure, the pharmacodynamic effects remained relatively unchanged. Inclisiran was found to be safe and well tolerated in patients with mild or moderate hepatic impairment.

This review focuses on the unique ADME characteristics of antisense oligonucleotide (ASO) drugs, which have diverse modes of drug actions and great potential for treating diseases untreatable by small molecule-based drugs. A meta-analysis of 10 FDA-approved ASO drugs provides comprehensive insights into their formulation, dosage, administration sites, distribution, metabolism, and excretion, improving our understanding of their therapeutic efficacy and potential adverse reactions.

Effect of ion-pairing reagent hydrophobicity on liquid chromatography and mass spectrometry analysis of oligonucleotides

Summary: We systematically studied the performance of thirteen alkylamines as ion-pairing reagents for ion-pair reversed-phase liquid chromatography (IP RP LC) and proposed a method to classify the hydrophobicity of alkylamines. The results showed that alkylamines with higher hydrophobicity allowed for effective separation of oligonucleotides at lower buffer concentrations, and replacing acetic acid with hexafluoroisopropanol improved the mass spectrometry signal.
