Anti-photobleaching cyanine-based nanoparticles with simultaneous PET and ACQ effects for improved tumor photothermal therapy

Summary: Designing anti-photobleaching photothermal agents (PTAs) is important for tumor photothermal therapy (PTT). Heptamethine cyanine (Cy7) dyes have gained attention in PTT, but their poor photostability limits clinical application. In this study, two new Cy7 dyes, DTZCy and WTZCy, were prepared and showed better anti-photobleaching performance compared to the FDA-approved ICG dye. The contributions of quenching fluorescence and enhancing photostability to improving the photothermal conversion efficiency of Cy7-based PTAs were discussed. DTZCy demonstrated higher PCE (eta = 31.8%) than WTZCy (eta = 16.5%) due to increased photostability caused by intramolecular photoinduced electron transfer (PET) effect. Silica nanoparticles encapsulating DTZCy, named DTZCy@SiO2 NPs, exhibited higher PCE (eta = 34.6%) and satisfying photo thermal stability both in vitro and in vivo, attributed to improved anti-photobleached ability caused by the aggregation-caused quenching (ACQ) mechanism. This work suggests new strategies for constructing anti-photobleaching PTAs by utilizing PET and ACQ mechanisms for efficient tumor PTT.
